摘 要:目的反相高效液相色谱法测定黄柏药材中黄柏碱和木兰花碱的含量。方法采用Phenomenex RP-C18色谱柱(4.6 mm×250 mm,5μm),乙腈-0.05 mol.L-1磷酸水溶液(10∶90,每100 ml加二乙胺0.2 ml)为流动相,流速1.0 ml.min-1,柱温30℃,检测波长225 nm。结果黄柏碱,木兰花碱分别在0.338 4~2.030 4μg,0.307 2~1.843 2μg范围内线性关系良好;平均回收率(n=6)分别为100.85%,100.35%。结论该方法为黄柏药材的质量控制提供实验依据,该方法具有简便、快速、准确、重现性好的特点。Objective To establish an RP - HPLC for determination of phellodendrine and magnoflorine in Cortex Phellodendri. Methods SymmetryShieldTM RP- C18 Column( 4.6 mm× 250 mm,5 μm ) was adopted;The mobile phase was acetonitrile - 0.05 mol L-1 phosphoric acid ( 10:90,0.2 ml diethylamine in every 100rnl ) at the flow rate of 1 ml min-l; The column temperature was 30 ~C and the detection wavelength was 225 nm. Results The linear ranges of phellodenrine and magnoflorine were 0. 338 4 - 2. 030 4 μg and 0. 307 2 - 1. 843 2μg;The average recoveries( n = 6) for the sample preparation of the markers were 100.85%, 100.35 %, respectively. Conclusion The quantitative method for quality control of phellodendrine and magnoflo- fine in Cortex Phellodendri by RP- HPLC provided the basis for quality coantrol of Cortex Phellodendri,this method is simple,rap- id and accurate with good reproducibility.
