摘 要:目的建立素馨花的总环烯醚萜苷提取物的质量标准。方法采用颜色反应和TLC(薄层色谱)对素馨花提取物中总环烯醚萜苷进行定性鉴别;用紫外分光光度法测定了有效部位总环烯醚萜苷的含量;用高效液相色谱法测定了橄榄苦苷的含量。结果在TLC图谱中可检出橄榄苦苷的特征斑点;建立了UV法测定总环烯醚萜苷含量的分析方法,总环烯醚萜苷的标准曲线为:A=0.028 9 C-0.004 9(R2=0.999 8),3.96~19.8 0μg/ml的范围内线性关系良好,平均回收率为102.64%,RSD=0.65%(n=6);建立了HPLC法测定橄榄苦苷含量的分析方法,橄榄苦苷的标准曲线为:Y=133 959X+23.44(R2=0.999 8),0.02~0.10 mg.ml-1范围内线性关系良好,平均回收率为100.12%,RSD=0.883%(n=6)。结论该方法稳定、准确、精密度高、专属性强、操作简便可行,适用于提取工艺和纯化工艺优化过程中的含量测定。Objective To establish the quality standards for total iridoid glycosides from Jasminum officinale L. var. grandiflo- rum. Methods Color reaction and TLC for qualitative identification of total iridoid glycosides from Jasminum officinale L. var. grandiflorum; Effective part of total iridoid glycosides content was determined by UV; Oleuropein content using HPLC. Results - Characteristic spots can be detected in the TLC maps of oleuropein; Established UV method for determination of total iridoid gly- coside content analysis method, the standard curve of total iridoid glycosides as follows : A = 0. 0289 C - O. 0049 (R2 = 0.999 8 ), 3.96 - 19.8 μg / ml linear in the rangegood relationship, the average recovery was 102.64%, RSD =0.65% (n = 6) ; Estab- lished HPLC determination of oleuropein content analysis method, oleuropein standard curve: Y = 133959X + 23.44 (R2 = 0.999 8) , 0.02 - 0.10mg ml-1 range a good linear relationship , the average recovery was 100.12% , RSD =0. 883% (n = 6). Conclusion The method is stable, accurate, high precision, strong specificity, operation is simple and feasible, is suitable for extraction and purification process optimization content determination.
