摘 要:目的研究槲皮苷对家兔离体主动脉环的作用,并探讨其可能的作用机制。方法采用家兔离体主动脉环灌流模型,观察累积浓度的槲皮苷对基础状态和去甲肾上腺素(NE)预收缩血管环的作用,并观察不同抑制剂对槲皮苷舒张血管作用的影响。结果槲皮苷(1×10-6~1×10-3μmol/L)对基础状态内皮完整血管环的张力无影响;对NE预收缩的血管环,槲皮苷在累积浓度(1×10-4~1×10-3μmol/L)时能剂量依赖性和非内皮依赖性的舒张血管环,并且该作用能被MB预处理所抑制,但不能被L-NAME和Indo预处理所抑制。结论槲皮苷(1×10-4~1×10-3μmol/L)对主动脉具有非内皮依赖性的舒张作用,其舒张机制可能是通过直接激活血管平滑肌的鸟苷酸循环。Objective To investigate the effect of Quercitrin on isolated rabbit aortic rings and the underlying mechanisms. Meth- ods Quercitrin was cumulatively added into organ bath to observe the isometric tension of thoracic aortic rings in basal tension or preconstricted by NE. Three inhibitors were used to explore the mechanism respectively. Results Cumulative administration of Quercitrin ( 1 ×10-6~1×10-3 μmol/L) did not affect the vasomotion of aortic rings with endothelium in basal tension; The rab- bit arteries with or without endothelium induced by NE could be dose - dependently relaxed by Quercitrin ( 1 ×10-4~1×10-3 μmol/L) ,which was inhibited by preincubation with MB ,but was not inhibited by preincubation with L -NAME and Indo. Con- clusion The results indicated that the endothelium - independent relaxation effect of Quercitrin might associated with cGMP.
