胰岛素样生长因子-Ⅱ及基质金属蛋白酶-9与早期自然流产的相关性研究

摘　　要：目的探讨胰岛素样生长因子-Ⅱ(IGF-Ⅱ)和基质金属蛋白酶-9(MMP-9)在早期自然流产发生机制中的作用以及二者的相关性。方法采用实时荧光定量PCR技术检测20例正常早孕妇女和30例不明原因早期自然流产患者绒毛组织中IGF-ⅡmRNA及MMP-9 mRNA的表达量;免疫组化检测IGF-Ⅱ及MMP-9的表达,并分析二者相关性。结果早期自然流产组IGF-ⅡmRNA和MMP-9 mRNA的表达水平明显低于对照组,差异有统计学意义(P<0.05)。早期自然流产组IGF-Ⅱ及MMP-9蛋白表达明显低于对照组(P<0.05),两组平均光密度(MOD)呈正相关,差异有统计学意义(r=0.491,P<0.05)。结论 IGF-Ⅱ及MMP-9蛋白参与早孕绒毛的发育,两者表达降低与早期自然流产密切相关,其机制可能为IGF-Ⅱ及MMP-9在绒毛中的表达下降,导致绒毛滋养细胞浸润能力不足,发生流产。Objective ： To investigate the expression of insulin-like growth factor- Ⅱ （ IGF- Ⅱ ） and matrix metalloprotein- ases-9（MMP-9） in the villi of women with early spontaneous abortion and discuss their role in the pathogenesis of early spontaneous abortion. Methods ： We selected 20 women in early nomal pregnancy and 30 women in early spontaneous abor- tion. The expression of IGF- Ⅱ and MMP-9 mRNA was detected by real-time fluorescence quantitative polymerase chain re- action （PCR）. At the same time,the expression and distribution of IGF-Ⅱ and MMP-9 protein in the villus of the two groups were observed with technique of immunohistochemical staining. Results： IGF-Ⅱ mRblA and MMP-9 mRNA were significantly less in the abortion group than those in the normal early pregnancy group （ P 〈 0.05 ）. Immtmo-histoehemistry analysis showed that the levels of IGF- II and MMP-9 were both significantly lower in the abortion group than those in the normal early pregnancy group. There was a positive correlation between them and the proportion was significantly different （ r = 0. 491, P 〈 0.05 ）. Conclusion： The protein of IGF- Ⅱ and MMP-9 participate in the development of the villi and their expressions are significantly lower, which is closely related to early spontaneous abortion, indicating that IGF- 11 and MMP- 9 may play an important role in the early spontaneous abortion.
