摘 要:目的:研究-格列美脲对高糖环境下大鼠下颌骨成骨细胞葡萄糖摄取的影响。方法:原代培养分离下颌骨成骨细胞并鉴定,实验分组:5.5mMG组(对照组);5.5mMG+格列美脲组;16.5mMG组;16.5mMG+格列美脲组,每组加入1ml含7.5μCi/ml18F-FDG的PBS,测定用总放射性计数与蛋白质浓度的比值。结果:高糖环境降低了成骨细胞糖摄取,格列美脲促进两种糖浓度细胞糖摄取。结论:格列美脲在成骨细胞的糖代谢中发挥重要作用。Objective: To explore the effect of glimepiride on glucose uptake of rat mandibular osteoblasts in hyperglycemia. Methods: Primary osteoblasts were isolated and cultured. Then the cells were placed in an osteogenic medium, containing different containing two concentrations of glucose (5.5mM and 16.5mM), with or without glimepiride (101.LM). Glucose uptake was detected by using ^18F-FDG in the cells. Results: The 16.5mM of glucose significantly inhibited ^18F-FDG uptake. The addition of glimepiride to the culture medium containing 5.5mM and 16.5mM glucose concentrations increased significantly glucose uptake. Conclusions: It was shown that glimepiride played an important role in glucose metabolism in osteoblasts.
