摘 要:目的观察柴胡对肝星状细胞c-fos和c-jun基因表达的影响。方法在培养的肝星状细胞株中分别加入不同浓度的柴胡(终质量浓度1.0、0.5g/L);于8、24、48、72h4个时间点分别收集细胞,提取肝星状细胞总RNA;用逆转录聚合酶链反应(RT.PCR)法测定c—fos和c-jun的基因表达水平。结果柴胡两组HSC—T细胞c.fos基因表达水平在8h(0.36±0.11、0.51±0.12)、24h(0.27±0.09、0.43-t-O.10)、48h(0.23±O.04、0.32±0.07)、72h(0.13±0.03、0.24±0.04)4个时间点均明显低于对照组(0.63±0.13、0.50±O.12、0.43±0.06、0.32±0.04);c-juII基因表达水平在8h(O.44±0.11、0,68±0.13)、24h(0.40±0.09、0.60±0.11)、48h(0.26±0.08、0.43±0.11)、72h(0.20±0.04、0.33±0.06)4个时间点也均明显低于对照组(0.93±0.13、0.80±0.12、0.63±O.10、0.46±0.07);随剂量加大,HSC—T细胞c—los、c-jun基因表达水平逐渐降低;两组间差异有统计学意义(P〈0.05)。结论柴胡对肝星状细胞c—fos和c-jun基因表达有明显的下调作用。Objective To investigate the effects of Radix bulpleuri on c-los and c-jun gene ex- pression in hepatic stellate cells. Methods Hepatic stellate cells (HSC-T6) were cultured in medium con- taining different concentrations of Radix Bulpleuri ( 1, O, and 0. 5 g/L) and harvested at different time points (8, 24, 48, and 72 h). Total RNA of HSC-T6 cells was isolated, and the expression levels of c-los and e-jun genes were detected by using reverse transcription polymerase chain reaction. Results The c-los gene expression levels of HSC-T6 celLs in two Radix Bulpleuri groups were significnatly lower than those in control group (0. 63 ±0. 13, O. 50 ±0. 12, 0. 43 ±0. 06, and O. 32 ±0.04) after exposure to Radix Bul- pleuri at8 h (0.36 +0. 11, 0.51 ±0. 12), 24 h (0.27 +0±09, 0.43 ±0. 10), 48 h (0.23 ±0.04, O. 32 +0. 07) and 72 h (0. 13 ±0. 03, 0. 24 ±0.04), The c-jun gene expression levels of HSC-T6 cells in two Radix Bulpleuri groups were also significantly lower than those in control group (0. 93 ± 0. 13, 0. 80 ± O. 12, 0:63 ±0. 10, and 0.46 ±0.07) after exposure to Radix Bulpleuri at 8 h (0.44 ±0. 11, 0.68 ± 0. 13), 24 h (0.40±0.09, 0.60±0. 11), 48 h (0.26±0.08, 0.43 ±0. 11) and72 h (0.20±0.04, 0. 33 ± 0.06 ). The c-fos and c-jun gene expression levels in HSC-T6 cells were decreased gradually as the dosage of Radix Bulpleufi increased, and there was significant difference in the c-los and c-jun gene ex- pression between two Radix Bulpleuri groups. Conclusion Radix Bulpleuri may down-regulate the c-los and c-jun gene expression in hepatic stellate cells.
