摘　　要：目的探讨肿瘤坏死因子-α（TNF-α）基因启动子区-308G/A多态性与潍坊地区汉族人群儿童支气管哮喘发病的相关性.方法应用PCR-RFLP方法对91例支气管哮喘患儿（哮喘组）和89例健康儿童（对照组）TNF-α基因-308G/A多态性进行检测.结果 TNF-基因-308G/A多态性,GG,GA,AA基因型频率哮喘组分别为83.5%,15.4%,1.1%,对照组分别为92.1%,7.9%,0（χ^2=3.54,P〉0.05）;G和A等位基因频率哮喘组为91.2%和8.8%,对照组为96.1%和3.9%（χ^2=3.55,P〉0.05）.与GG基因型个体比较,GA基因型携带者儿童发生哮喘的风险增加2.16倍（95%CI：1.94-5.55）,与G等位基因个体比较,A等位基因携带者儿童发生哮喘的风险显著增加2.35倍（95%CI：2.31-5.71）.结论 TNF-基因-308G/A多态性与潍坊地区汉族人群儿童支气管哮喘病的发生无明显相关性.Objective To investigate the association of tumor necrosis factor-α（TNF-α） gene promoter region-308 G/A polymor-phism with bronchial asthma of children in Weifang Han population .Methods The TNF-α-308G/A polymorphism was determined by PCR-RFLP in 91 patients with asthma（asthma group） and 89 normal children（control group）.Results The frequencies of GG,GA and AA geno-types of TNF-α-308G/A polymorphism were,respectively,83.5%,15.4%and 1.1%in asthma group,92.1%,7.9%and 0 in control group （χ^2 =3.54,P〉0.05）.The G and A allele frequencies of TNF-α-308G/A polymorphism were,respectively,91.2% and 8.8% in asthma group,96.1%and 3.9%in control group（χ^2 =3.55,P〉0.05）.No significant differences were found in allele and genotype frequencies of TNF-α-308G/A between the two groups.The individuals with GA genotype and A allele were 2.16（95%CI：1.94~5.55） （P〉0.05） and 2.35（95%CI：2.31~5.71） （P〉0.05） times susceptible to asthma than the normal controls ,compared with GG genotype and G allele . Conclusion These results indicated that TNF-α-308G/A polymorphism did not show association with bronchial asthma of children in Wei -fang population.
