摘  要:耐热芽孢菌污染是目前困扰着乳品企业的一大难题。从奶粉样中分离得到一种极端嗜热菌,命名为KLDS9.1101,其可耐100℃、10min的高温条件。通过有效地筛选分离,并采用PCR扩增、琼脂糖凝胶电泳、16SrDNA等分子生物学方法,鉴定了该菌为好热黄无氧芽孢菌,并构建了系统发育树,确定其种属关系。此外还以好热黄无氧芽孢菌为实验菌株,探寻利用过氧乙酸消毒耐热芽孢菌的方法,得知在200mg/L浓度下作用3min以上可以基本对好热黄无氧芽孢菌消毒,为乳品工厂中耐热芽孢菌控制提供理论基础。Thermophilic bacillis is a big problem puzzling dairy factory. An extremLy thermophilic microrganism named KLDS 9.1101 which can survive from 100 ~, 10 min, was isolated from milk powder. By the use of PCR amplification, agarose gel electrophoresis, 16SrDNA and other molecular biological methods, this bacteria was identified as Anoxybacillus flavithermus. Meanwhile, a phylogenetic tree was also constructed to make their relationship clear. In addition, an effective method was developed to kill the bacteria by the use of peracetic acid. It was concluded that Anoxybacillus flavithermus cells can be mostly disinfected in peracetic acid longer than 3 min, at the concentration of 200 mg/L. It can provide theoretical basis to control thermophilic bacillis in milk powder factory.
