摘  要:利用cDNA-AFLP技术,鉴定了1个对低磷胁迫逆境产生明显应答的小麦bHLH型转录因子转录本片段(TDF)。比对分析表明,该TDF(TaHLH1 EST)与前人在水稻中鉴定的对低磷产生明显应答的bHLH型转录因子基因OsPTF1高度同源。利用生物信息学技术,获得了TaHLH1 EST对应的cDNA序列。TaHLH1的cDNA全长为2 209bp,含有1个编码480个氨基酸残基的开放阅读框。TaHLH1含有bHLH型转录因子具有的Basic-Helix-Loop-Helix保守基序,分别由13,15,6和22个氨基酸残基组成。比对分析表明,TaHLH1与源于水稻、大麦和玉米的同源基因编码蛋白在保守基序的氨基酸组成上高度同源。TaHLH1对低磷、低氮、干旱和高盐等逆境明显应答,但对低钾、低钙、脱落酸(ABA)和低温不产生应答。研究表明,TaHLH1是小麦bHLH型转录因子家族的重要成员,在介导低磷等非生物逆境的信号转导中可能发挥着重要作用。Using cDNA-AFLP technique, a transcript-derived fragment (TDF) classified into bHLH type transcription factor family that responds to low-Pi stress was identified. Alignment analysis showed that this TDF (TaHLH1 EST) shares a high similarity to OsPTF1, a bHLH type transcription factor gene in rice that responds also to the low-Pi stress. Based on bioinfor- matic analysis, the cDNA sequence corresponding to TaHLH1 EST was obtained. TabHLH1 has 2 209 bp-long of full length cDNA and encodes a 480 aa-long of polypeptide. TahHLH1 contains the conserved Basic-Helix-Loop-Helix motif, with the amino acid residues of 13,15,6, and 22, respectively. Alignment analysis revealed that the conserved domains among TaHLH1 and its homologs of rice, barley, and maize are highly identical. The expression level of TaHLH1 was induced by several abiotic stresses, including low-Pi, low-nitrogen, drought, and high salinity, but was not responded to stresses of low-K, low-Ca, abiscisic acid (ABA),and low temperature. The results in this study suggest that TaHLH1 is one member of bHLH type transcription factor family in wheat and possibly plays an important role on mediation the transduetion of signals initiated by stresses of low-P, low-N, drought, and high salinity.
