摘  要:目的:探讨Wnt10b对体外长期培养的人毛乳头细胞诱导毛囊生成能力的影响作用。方法:将人毛乳头细胞在体外传代培养到第8代后,加入人重组蛋白Wnt10b共培养,通过免疫荧光染色检测人毛乳头细胞的β-catenin表达,Real-time QPCR检测与毛乳头细胞诱导能力相关的特异性标记物碱性磷酸酶(ALP)和胰岛素样生长因子-1(IGF-1)的表达,以及ALP染色检测Wnt10b对人毛乳头细胞的作用。结果:Wnt10b能够通过经典的Wnt/β-catenin信号通路作用于体外长期培养的人毛乳头细胞,上调人毛乳头细胞特异性标记物ALP和IGF-1的表达,从而维持人毛乳头细胞的诱导能力。结论:Wnt10b能够通过上调特异性标记物ALP和IGF-1的表达来维持体外长期培养的人毛乳头细胞的毛囊诱导能力。Objective To investigate the effects of Wnt10b on the hair follicle induction of long-term cultured human dermal papilla cells (HDPCs) in vitro. Methods The HDPCs of generation ability 8th were used for experiments. The recombinant human WntlOb (1 μg/mL) was added to the medium and cultured with HDPCs. To evaluate the effects of WntlOb treatment, β-eatenin expression and alkaline phosphatase (ALP) activity of the HDPCs were examed, and the gene expression of HDPCs markers were determined by Real-time quantitative PCR. Results WntlOb has a significantly effects on the HDPCs through Wnt/β-catenin signaling pathway, it can successfully upregulated the specific expression of HDPCs hair follicle induction marker molecules, such as ALP and insulin-like growth factor-1 (IGF-1). Conclusion WntlOb can maintain the hiar follicle-inducing ability of the long-term cultured HDPCs through the classic canonical Wnt/β-catenin signaling pathway in vitro.
