山莨菪碱预防大鼠肺水肿时动脉血浆与支气管肺泡洗涤液中PGE_2和LTC_4含量的变化

摘　　要：目的　通过研究山莨菪碱 (ADM ,6 5 4 2 )预防大鼠肺水肿时 ,对动脉血浆和支气管肺泡洗涤液 (BAL)中的PGE2和LTC4 含量的影响 ,探讨山莨菪碱防治肺水肿的部分机制。方法　先从股静脉注射山莨菪碱 (30mg·kg-1)作为预防 ,再注射肾上腺素 (0 .3mg·kg-1) ,30min后收集动脉血浆和BAL ,用放射免疫法分别测定正常对照组、肺水肿组、山莨菪碱预防组的PGE2 和LTC4 的含量。结果　肺水肿组动脉血浆中的PGE2 和LTC4 的含量、LTC4 与PGE2 比值高于正常对照组 (P <0 0 1) ,BAL中的PGE2 含量低于正常对照组 (P <0 0 5 ) ,LTC4 的含量、LTC4 与PGE2 比值高于正常对照组 (P <0 0 1) ;山莨菪碱预防组动脉血浆和BAL中PGE2和LTC4 含量、LTC4 与PGE2 比值均低于肺水肿组 (P <0 0 1)。结论　山莨菪碱能抑制肺水肿时PGE2 和LTC4 的释放 ,从而改善血管的通透性 。AIM To investigate the effect of anisodamine (654 2) on PGE 2 、LTC 4 content and ratio of LTC 4 to PGE 2 of arterial plasma and BAL in rats with pulmonary edema.METHODS\ Pulmonary edema (PE) similar to the neurogenic type was induced by adrenaline(AD 0 3 mg·kg -1 ,iv) in rats.In 654 2 preventive group the 654 2 (30 mg·kg -1 iv)was given before AD administration. PGE 2 and LTC 4 level in plasma and BAL were estimated by RIA. RESULTS 654 2 could inhibit the increase ofPGE 2、LTC 4 and modulating PGE 2 and LTC 4 to normal level in plasma and BAL. CONCLUSION\ 654 2 may inhibit arachidonic acid metabolism by during pulmonary edema produced.It may be one of the preventive mechanisms against pulmonary edema.
