摘 要:以陆地棉种质系珂字棉312为材料,探讨了在共培养基添加乙酰丁香酮(AS)同时调节pH值对抗性愈伤组织诱导率、正常体细胞胚数以及Kan抗性苗获得率的影响。结果表明,在共培养阶段的乙酰丁香酮浓度与pH均对棉花遗传转化有明显的影响。当乙酰丁香酮的浓度为100μmol/L、pH值为5.6时,棉花的遗传转化率最高。其抗性愈伤组织的诱导率达到72.96%,Kan抗性苗获得率可达到8.96%;对转化再生植株进行PCR检测,约52.07%的Kan抗性苗呈阳性。Using hypocotyl of Coker 312 (Gossypium hirsuturnL. ) as the experimental material,this study investigated the effect of acetosyringone (AS) concentration and pH in the co-culture medium on the resistant calli-inducing rate,number of somatic embryos and transformation efficiency. The results showed that the transformation efficiency was significantly affected by acetosyringone (AS) concentration and pH in the co-culture. The optimized acetosyringone concentration was 100μmol/L and the appropriate pH was 5.6, which can significantly improve the efficiency of genetic transformation in cotton. The rate of resistant calli was 72.96% and the harvesting proportion of Kan+ regenerated plants reached 8. 960/oo. Regenerated plants were confirmed by PCR and about 52.07% regenerated plants were proven to be resistant.
