中医不同治法对骨质疏松症大鼠骨钙素mRNA和蛋白表达影响的比较研究

摘　　要：目的:观察中医不同治法对糖皮质激素诱导骨质疏松症大鼠骨钙素mRNA和蛋白表达的影响,探讨中医防治骨质疏松症的作用机制。方法:将120只雌雄各半的大鼠随机分为正常对照组、模型对照组、补肾中药组、健脾中药组、活血化瘀中药组和骨疏康中药组6个组。用地塞米松肌注造模。实验结束后,腹主动脉取血处死大鼠,用PCR法及Western印迹法检测各组大鼠骨组织骨钙素mRNA和蛋白表达。结果:①与正常组相比,模空组大鼠骨组织骨钙素mRNA表达水平明显下降(P<0.05),与模空组相比,各治疗组大鼠骨钙素mRNA表达水平均有不同程度的升高,其中补肾组升高趋势最为显著(P<0.01)。②与正常组比较,模空组大鼠骨钙素的蛋白表达明显降低,但统计比较无显著差异;与模空组比较,各治疗组大鼠骨钙素的蛋白表达均明显升高,其中补肾组升高趋势最显著(P<0.01)。结论:补肾方法通过调节骨钙素对骨质疏松症具有一定的防治作用。Objective：To approach the effect of different Chinese medicine treatments on the mRNA and protein expression of bone gla protein in bone tissue in GC-induced osteoporosis.Method：120 rats（male/female=1∶ 1）were randomly divided into 6 groups：blank controlling group,model controlling group,invigorating the kidney group,invigorating the spleen group,promoting blood flow group and Gusukang group.RT-PCR method was used to examine the mRNA and protein expression of bone gla protein in the skeleton and skeletal muscle.Result：①Compared with normal group,bone gla protein mRNA expression in bones of blank model group decreased significantly（P〈0.05）;compared with blank model group,bone gla protein mRNA expression of all the treating groups increased differently,among which reinforcing the kidney group increased most significantly（P〈0.01）.② Compared with normal group,bone gla protein expression in bones of blank model decreased,but made no scense in statistic;compared with blank model group,bone gla protein expression of all the treating groups increased,among which reinforcing the kidney group increased most significantly（P〈0.01）.Conclusion：To some degree,invigorating the kidney can prevent and treat osteoporosis by regulating bone gla protein.
