摘  要:目的:研究大黄酸对大鼠非酒精性脂肪肝(NAFLD)的防治作用及其部分机制。方法:采用高脂饲料12周喂养建立大鼠NAFLD模型;50只SD大鼠随机分为5组:正常对照组、模型组、大黄酸低、中、高剂量[100,200,400 mg/(kg.d)]组(每组10只)。给药4周后处死大鼠,计算肝脏指数,观察肝组织病理形态学变化,全自动生化仪检测血清ALT、AST、FFA、LDL-C、HDL-C、TG和TC含量;比色法测定肝组织MDA、GSH-PX水平;Real-time PCR检测肝脏组织PPAR-γmRNA表达。结果:与模型组比较,大黄酸给药可使大鼠肝组织脂肪变性明显减轻,肝脏指数明显降低;各给药组血清ALT、AST、FFA、TG、TC和肝脏MDA水平降低,肝脏GSH-PX活力升高(P<0.05或0.01);模型组肝脏PPAR-γmRNA表达明显升高(P<0.01),大黄酸干预组则明显降低其表达(P<0.01)。结论:大黄酸对大鼠NAFLD具有较好的防治作用,其机制可能与清除氧自由基、减少脂质过氧化产物和改善脂质代谢紊乱有关。Objective:To investigate the beneficial effects of Rhein on nonalcoholic fatty liver disease(NAFLD)in rats fed with high fat diet and to explore the possible mechanisms.Methods:50 SD rats were divided into normal group,control group,and low,medium,high dose of Rhein [100,200,400 mg/(kg·d),respectively]group randomly(10 rats in each group).NAFLD was induced by a formula of high fat(HF)diet.At the treatment time of 4 weeks later,rats were sacrificed,and hepatic index was calculated.Histologic changes were observed and compared between these groups.Serum levels of ALT,AST,free fatty acids(FFA),LDL-C,HDL-C,TG and total cholesterol(TC)were detected by automatic biochemical analyzer,hepatic contents of malondialdehyde(MDA)and glutathione peroxidase(GSH-PX)were evaluated by colorimetric technique,hepatic expression of peroxisome proliferator-activated receptor(PPAR)-γ mRNA was detected by real-time PCR.Results:Comparing with that of control group,liver steatosis in those Rhein groups were alleviated obviously,and hepatic indexes were decreased also.A consistent decline of serum levels of ALT,AST,FFA,LDL-C,TG,TC and hepatic MDA,and increase of hepatic GSH-PX were obtained in all of the three Rhein groups(P〈0.05 or P〈0.01).As for the hepatic expression of PPAR-γ mRNA,HF diet resulted in a marked increase but improved effectively by Rhein(P〈0.01 for all).Conclusions:Rhein improves NAFLD in a rat model of HF diet,the effect is contributed partly to the mitigated oxidative stress and improvement of lipid metabolism.
