摘  要:目的:探讨大剂量昆明山海棠对大鼠睾丸的毒性损害及其可逆性。方法:36只成熟雄性SD大鼠,随机分成2组,每组18只:对照组喂服生理盐水10 mL.kg-1.d-1,给药组喂服昆明山海棠22.5 g.kg-1.d-1。连续60 d后,随机处死半数大鼠,取材。余下大鼠恢复饲养72 d后,处死取材。检测睾丸系数、睾丸体积、精子密度、形态、活力等。光镜观察睾丸组织的病理变化。结果:给药60 d,与对照组比较,给药组睾丸系数、精子密度显著下降(P<0.01)。给药组精子形态异常,活动力丧失,光镜下睾丸组织严重损伤。停药72 d后,睾丸毒性损害无恢复。结论:大剂量昆明山海棠具有明显的睾丸毒性,且毒性损害难以恢复。Objective:To study anti -reproductive effects of Tripterygium Hypoglaucum Hutch (THH), and whether the effects are reversible at a large dose on testicle tissue in male rats. Methods :36 SD male rats were randomly divided into 2 groups,18 in each group. The control group were given normal saline at a dose of 10 mL ·kg^-1·d^-1 ,while the experi- ment group were given THH at a dose of 22.5 g ·kg^-1·d^-1. After 60 days, half rats of each group were sacrificed randomly. The rest rats were continually raised until 72 days later, then sacrificed. The relative testis weight, testicle volume, sperm density and the total activity were comparatively analyzed. The pathological changes in testieular tissue were observed by optical microscope. Results:Compared with the control group, a significant decrease in relative testis weight and sperm density were seen in the experiment group after a 60 - day treatment( P 〈 0.01 ). In experiment group the motility of sperm from caudal epididymides dropped to zero, and diastrophic sperms were seen. The testicular lesion in rats caused by THH was serious. It was not found that epididymal lesion recovered after 72 days treatment. Conclusion : It indicats that for male rats,THH has obvious and irreversible toxic effect on the testicular tissue.
