解毒化瘀颗粒对术后肝衰竭大鼠肝细胞再生的促进作用

摘　　要：目的探讨解毒化瘀颗粒对大部分肝切除肝衰竭大鼠肝细胞再生的的促进作用。方法健康SPF级Wistar大鼠300只随机分为6组:假手术组50只、对照组50只、解毒化瘀颗粒组50只、解毒组50只、化瘀组50只、祛痰组50只。采用85%肝叶切除术构建大鼠肝衰竭模型,秤取肝脏重量,计算肝再生指数,使用流式细胞仪进行检测,每份样品检测104个细胞,应用DNA细胞周期分布软件计算出肝细胞增殖指数。结果在术后48h之后解毒化瘀颗粒组存活率显著优于对照组(P<0.05);解毒化瘀颗粒组在72h、168h时肝脏再生指数与对照组比较有统计学差异(P<0.05);解毒化瘀颗粒组PI峰值出现在术后24h、48h,与对照组有显著性差异(P<0.05)。结论解毒化瘀颗粒可促进肝细胞再生,提高术后肝衰竭大鼠存活率。ObjectiveStudy of Jiedu Huayu granules on most of the liver resection of liver failure in rat liver cell regeneration promoting effect. MethodsHealth level SPF 300 Wistar rats were randomly divided into 6 groups： sham operation group was 50,50 in control group,detoxification and dissipation blood stasis granule group 50,group 50,group of detoxification Huayu 50,and group 50.The construction of 85% hepatectomy rat liver failure model,scale calculation of liver weight,liver regeneration index,using flow cytometry for detection,each sample detection of 104 cells,the application of DNA cell cycle distribution software to calculate the liver cell proliferation index. ResultsIn the postoperative 48h Jiedu Huayu granules group survival rate was better than the control group（P0.05）;detoxification Huayu granule group at 72h,168h liver regeneration index significantly compared with the control group（P0.05）;Jiedu Huayu granules group PI peaks appear at 24h after operation,48h,and control group was significant difference（P0.05）. ConclusionJiedu Huayu granule can promote the regeneration of liver cells,can improve the postoperative liver failure on survival rate of rats.
