摘　　要：目的观察针叶聚戊烯醇对大鼠慢性免疫性肝损伤致肝纤维化的治疗作用。方法大鼠腹腔给予猪血清0.5 ml,每周两次,建立慢性免疫性肝损伤模型。造模同时开始给药,给药组灌胃给予低、中、高剂量的针叶聚戊烯醇(10,20,40 mg.kg-1),模型组给予等体积生理盐水,每日给药1次,连续8周。末次给药后,动物禁食12 h,颈动脉采血,离心,取血清进行肝功能指标、氧化损伤指标及肝纤维化指标检测;无菌取脾,进行淋巴细胞增殖能力检测;各组取肝脏同一部位做病理组织学检查。结果针叶聚戊烯醇中、高剂量组可以明显降低肝损伤大鼠血清透明质酸、IV型胶原和丙二醛水平以及脾淋巴细胞增殖能力,显著升高血清超氧化物歧化酶水平,有效改善肝功能指标和肝组织形态(P<0.05,P<0.01)。结论针叶聚戊烯醇对大鼠慢性免疫性肝损伤导致的肝纤维化有明显治疗作用,有望作为抗肝纤维化的新药进一步研发。ObjectiveTo investigate the therapeutic effects of polyprenols from pine needles on liver fibrosis induced by chronic immune liver injury in rats. MethodsRats were intraperitoneally administered 0.5 ml porcine serum twice a week to establish the chronic immunological hepatic injury model.The administration group was treated with polyprenols（10,20 and 40 mg·kg-1） and model group was treated with normal saline once a day for 8 weeks.Fasting for 12 hours after last administration,the blood from the carotid artery was collected and centrifugated,the serum was used for detection of liver function,oxidative damage as well as the indexes of hepatic fibrosis.The spleen was used for measurement of lymphocyte proliferation.Taking the same part of livers from each group to examine the liver histopathology. ResultsMiddle and high-dose of polyprenols markedly decreased the level of hyaluronic acid,collagen type IV,malondialdehyde and lymphocyte proliferation,significantly increased the level of superoxide dismutase,effectively improved the liver function and liver histology（P0.05 and 0.01）. Conclusionpolyprenols possess significant therapeutic effects on liver fibrosis induced by chronic immune liver injury and could get further development.
