摘 要:目的使用细菌氧化还原反应实验模型,以电极、计算机的组合技术,快速诊断VTEC O157的药物敏感性。方法 VTEC O157经抗生素处理后,将其附着在工作电极表面,在工作电极与对极之间以10 mV/s的递增速率输入阶梯形递增电压,计算机同步记录电压在0.00~1.00 V范围内的伏安图谱。根据伏安图谱上的峰电流值,判断VTEC O157对抗生素的敏感性;峰电流值≤2.5μA为敏感(S),峰电流值≥3.0μA为耐药(R),峰电流值2.6~2.9μA为中敏(M)。对照组采用常规琼脂扩散法诊断VTEC O157的药物敏感性。结果采用电极法,伏安图谱的记录及相关峰电流值数据的测定可在2 min内完成。本研究使用的VTEC O157菌株对13种抗生素的药敏试验结果表明,对Penicillin及Lincomycin二种抗生素不敏感,对Ampicillin及Erythromycin中度敏感,对余下的9种抗生素敏感。其结果同采用琼脂扩散法的结果一致。结论根据伏安图谱中峰电流值的大小,可间接判断VTEC O157对抗生素的敏感性。电极法的峰电流值与琼脂扩散法的抑菌圈直径成反比。Objective Using experimental model of bacterial oxidation reduction, the author rapidly diagnosed the sensitivity of VTEC O157 by combining technology of electrode and computer. Method After antibiotic treatment, the drug was adhered to the surface of the working electrode and a voltage with 10 mV/s increasing rate was input between the working electrode and the counter electrode. The computer simultaneously recorded the voltammogram of voltage in the range of 0.00-1. 00 V. Determined the sensitivity of VTEC O157 to antibiotic according to the value of peak current on the voltammogram. If the peak current value is less than or equal to 2.5 μA, it is sensitive(S) ; if the peak current value is more than or equal to 3.0 μA, it is resistant(R). And if the peak current value is of 2.6-2.9 μA, it is moderately sensitive(M). Agar diffusion method was used to diagnose drug sensitivity of VTEC O157 in the control group. Findings The electrode method can complete the recording of vohammogram and the relative peak current value measurement in two minutes. The testing of VTEC O157 strains with 13 kinds of antibiotics showed that: VTEC O157 was insensitive to Penicillin and Lincomycin, moderately sensitive to Ampicillin and Erythromycin, and sensitive to the remaining nine antibiotics. The results are consistent with those obtained by the agar diffusion method. Conclusion The sensitivity of VTEC O157 to antibiotic can be indirectly determined according to the value of voltammogram peak current on vohammogram. Peak current value of the electrode method is inversely proportional to the diameter of inhibition zone by agar diffusion method.
