摘  要:猪流行性腹泻(PED)是由猪流行性腹泻病毒(PEDV)引起的以腹泻、呕吐、脱水和哺乳仔猪高死亡率为特征的一种肠道传染病,给养猪业造成严重的损失。将构建的PEDV重组乳酸菌,诱导表达后,经不同免疫方式接种4周龄小鼠,1周后采血应用ELISA方法检测血清中的PEDV抗体及IFN-γ,评价PEDV乳酸菌疫苗刺激产生的体液免疫与细胞免疫水平。同时,在免疫前后测定小鼠体重,评价疫苗对小鼠生长发育的影响。结果显示,各免疫组对小鼠体重增加没有负影响,且略有增加;免疫后,10倍稀释口服组小鼠刺激产生的PEDV抗体水平最高,同时该组刺激产生的血清IFN-γ水平也最高。该PEDV乳酸菌载体口服疫苗能够刺激小鼠产生体液免疫和一定的细胞免疫。The porcine epidemic diarrhea (PED) is a highly contagious enteric disease in pigs causing vomit, diarrhea, dehydration and high mortality for piglets, and causes great loss in animal husbandry. In this research, the previous constructed recombinant Lactococcus expressing vector was used to challenge mouse at four weeks old by different inoculating approach after induction of the recombinant Lactococcus. A week later, the sera of mice in different groups were used to detect the specific PEDV antibody and IFN-7 by ELISA, and the degree of humoral immunity and cellular immunity induced by the recombinant Lactococ- cus vaccine was evaluated. Simultaneously, the body weights of mice in different groups were weighed to e- valuate the influence on the growth of mice by the recombinant Lactococcus vaccine. The results showed that the recombinant Lactococcus vaccine had no negative effect on the weight gain for the immunized groups of mice. Furthermore, a little increasing was shown in most of immunized groups of mice. After immunization, the groups immunized orally with 10 times of recombinant Lactococcus vaccine displayed the highest antibody titer and highest IFN-γ degree in sera detected by ELISA. The recombinant Lactococcus vaccine immunized orally can induce humoral immunity and cellular immunity at some extent in mice.
