摘 要:为研究德国牧羊犬嗅觉受体(OR)基因多态性,采用PCR技术对警用德国牧羊犬cOR52A17基因进行扩增、T-A克隆、测序、序列比对分析及转录因子预测。结果显示,德国牧羊犬cOR52A17基因启动子区域-114位点碱基发生替换(A>C);此突变导致cOR52A17基因启动子(-118bp~-113bp)区域成为转录因子AML-1a识别位点。研究表明,德国牧羊犬嗅觉受体基因具有多态性,且某些多态位点可能影响嗅觉受体基因的功能。SNP A-114C位点对cOR52A17基因功能的调控有待深入研究。To study polymorphism of olfactory receptor gene in German shepherd dogs, cOR52A17 gene was amplified and cloned through PCR and T-A cloning. The recombinant plasmids were sequenced and compared with the published sequences of dogs in GenBank. The result showed there was one mutation (SNP A-114C) in the promoter which induced to form the identifying sequence of transcription factor AML-la in region -118 bp to -113 bp. This suggested that olfactory receptor gene be polymorphisrh which might affect function of genes. Further research should be done to reveal how SNP A-114C regulted the function of cOR52A17 gene.
