摘 要:使用BellCell-500AP生物反应器和BioNOC○R-Ⅱ微载体,对BHK-21细胞在此新型高密度培养生物反应器中的生长特性进行研究。将含有1.5×108个BHK-21细胞的细胞悬液加入含有微载体的生物反应器内,待细胞贴附后加入细胞生长液,每日取载体计算细胞总数,观察BHK-21细胞的生长特性,并分析葡萄糖消耗规律。经过6d培养载体中细胞总数达到峰值,细胞密度是起始接入数量的31倍(4.41×109个),并且葡萄糖日消耗量与细胞增殖速度具有一定的相关性。试验成功实现了BHK-21细胞的高密度培养,为以BHK-21细胞为生产基质的病毒疫苗生产提供了一条简便快捷的途径。The paper focused on the growth characteristics of BHK-21 cells in a microcarrier(BioNOCC-Ⅱ) using a novel high-density cultivation oscillating bioreactor ( Bell Cell-500AP ). 1. 5×10^8 BHK-21 cells were inoculated into the bioreactor with microcarrier,then new cell growth medium was refilled after cells attached to the microcarrier. Growth characteristics and cells number of BHK-21 cells were observed each day, and glucose consumption rates in the medium were monitored daily. The results showed that BHK-21 cells reached a peak with 4. 41×10^9 on the 6 day post infection, which was 31 times higher than that in the initial inoculated cells, and glucose consumption each day was associated with cell proliferation rates. This study successfully achieved a high density culture for the BHK-21 cells, which would provide a simple and efficient way for the production of virus vaccine using the BHK-21 cells.
