摘  要:为了使临床合理使用抗菌药物,防止细菌耐药性的产生,采用琼脂倍比稀释法和肉汤富集法测定了5种氟喹诺酮药物对56株猪沙门菌的体外最小抑菌浓度(MIC)和防耐药变异浓度(MPC),计算MIC90、MPC90、MPC/MIC,结合药代动力学,比较5种氟喹诺酮类药物对猪沙门菌突变选择窗(MSW)的影响。试验结果显示,5种药物对猪沙门菌的MPC90的顺序为环丙沙星>恩诺沙星>吉米沙星=左氧沙星>加替沙星。结合药动学参数按规定剂量给药,血药浓度大于MIC90的时间顺序为加替沙星>左氧沙星>吉米沙星>恩诺沙星>环丙沙星,血药浓度大于MPC90的时间,加替沙星为24h,左氧沙星为6h,其他3种均达不到MPC90值。加替沙星和左氧沙星的防耐药突变能力强于其他3种喹诺酮药物,在临床治疗猪沙门菌感染时可作为首选药物。To develop strategies for reducing drug resistance development and provide rational application guidelines for available antibacterial medicines in clinic practice, the minimal inhibitory concentrations (MIC) and mutant prevention concentrations (MPC) of ciprofloxacin, levofloxacin, gatifloxacin, enrofloxa- cin and Gemifloxacin were determined in 56 strains of Salmonella isolated from swine by agar two-fold di- lution method. The values of MICg0, MPC00 and MPC/MIC were calculated. The result showed that the order of MPC90 was ciprofloxacin〉enrofloxacin〉gemifloxacin = levofloxacin〉gatifloxacin. Based on pub-lished data for the various fluoroquinolone pharmacokinetic parameters, the time (T) of the serum drug concentrations remained above the MICg0 in a rank order as follows: gatifloxacin〉levofloxacin〉gemifloxa- cin〉enrofloxacin〉ciprofloxacin. For T〉MPC90, gatifloxacin would be approximately 24 h and levofloxa- cin would be approximately 6 h. But other three fluoroquinolones did not exceed the MPC90. The ability of mutant prevention concentration of gatifloxacin and levofloxacin were superior than that of other three fluoroquinolones, which can be firstly selected for treating Salmonella infection.
