摘 要:目的研究新疆地区哈萨克族心房颤动与钾离子通道KCNE1-G38S基因多态性的相关性。方法收集新疆地区哈萨克族房颤人群(房颤组)和非房颤人群(对照组)各100例的外周血样标本,提取DNA,经聚合酶链反应-限制性内切酶片段长度多态性(PCR-RFLP)、琼脂糖电泳法鉴定分析KCNE1-G38S基因型和等位基因分布。结果KCNE1-G38S基因AA、AG、GG基因型频率在房颤组分别为0.13(13/100)、0.34(34/100)和0.53(53/100),在对照组分别为0.10(10/100)、0.57(57/100)和0.33(33/100),两组基因型分布有显著性差异(P<0.05),且房颤组G等位基因频率(0.70)明显高于对照组(0.515)(P<0.001)。结论新疆地区哈萨克族心房颤动的发生与KCNE1-G38S基因多态性相关,KCNE1-G38S基因多态性位点可能存在种族、地区差异。Objective To study if Xinjiang Kazak people's atrial fibrillation is related with potassium channels KCNE1-G38S gene polymorphisms. Methods The blood was Collected from Kazak people in xinjiang region with atrial fibrillation (af) and control group, both included 100 cases. DNA was extracted, polymerase chain reaction- restriction enzyme fragment length polymorphism (PCR-RFLP) was used and identified by agarose electrophoresis to analyse KCNE1-G38S genotype and allele distribution. Results KCNE1-G38S gene AA, AG, GG genotype fre- quency were found respectively in atrial fibrillation group 0. 13 ( 13/100 ), 0. 34 ( 34/100 ) and 0.53 ( 53/100), and in the control group 0. 10( 10/100 ), 0. 57 (57/100) and 0.33 (33/100), two groups of genotype distribution were statistically significant differences ( P 〈 0.05 ), and atrial fibrillation group G allele frequency (0. 70 ) was much higher than those in the control group (0. 515 ) ( P 〈 0. 001 ). Conclusions In Xinjiang Kazak people the occurrence of af is related with KCNEl-G38S gene polymorphism is related, and which may have race, regional differences.
