摘 要:目的:探讨人脐带间充质干细胞(human umbilical cord mesenchymal stem cells,hUCMSCs)在脊髓损伤(SCI)后减轻神经功能缺损的疗效,其对内源性细胞增殖的潜在影响。方法:雄性成年SD大鼠46只,以改良Allen′s法制备脊髓T9节段挫伤模型,用评估后肢的运动功能,HE染色、免疫组化染色和图像分析法观察GFAP,Nestin的表达情况。结果:hUCMSCs组在移植后行为学评分(BBB)明显高于PBS组(P<0.05),且损伤腔隙比PBS组更小(P<0.05);移植组中BrdU阳性细胞比对照组增加显著。结论:hUCMSC促进内源性细胞的增殖,有助于脊髓损伤后功能恢复。Objective To investigate the efficacy of hman umbilical cord mesenchymal stein (:ells (hUCMSCs) in reducing neurological deficits alter spinal cord injury (SCI), and to explore its potential impael on the proliferation of endogenous cells. Methods 60 male SD rats were randomly divided into control group (PBS group) and hUCMSCs transplantation group (hUCMSCs group). The SC1 animal model was induced by new Allen's method. The motor function of hind limbs was evaluated by BBB scores. The expressions of (;FAP and Nestiu were measured by HE staining, immunohistochemieal staining, and imaging analysis. Results Alter transplantation, BBB scores of hUCMSCs gruup were significantly higher than those of PBS group (P 〈 0.05), damage cavities were smaller than those of PBS group (P 〈 0.05), and BrdU positive cells were increased significantly as compared to PBS group (P 〈 0.05). Conclusions hUCMSC could promote the proliferation of endogenous cells, and contribute to the functional recovery after spinal cord injury.
