摘  要:目的:探讨活血药(当归、川芎)、破血药(三棱、莪术)对动脉粥样硬化(AS)大鼠主动脉内膜B细胞白血病-淋巴瘤-2(Bcell leukemia-lymphoma-2,Bcl-2)、B细胞白血病-淋巴瘤-2相关蛋白基因(Bcl-2-associated X proteingene,Bax)表达及细胞凋亡率的影响。方法:将60只雄性Wistar大鼠随机分为5组,即空白组、模型组、活血药(当归、川芎)组、破血药(三棱、莪术)组、他汀组,每组12只。先采用高脂喂饲复制AS大鼠模型,造模成功后,灌胃给药,连续4周。实验结束时检测Bcl-2、Bax蛋白表达及细胞凋亡率。结果:他汀组、破血药组Bcl-2表达明显高于模型组(P<0.01);他汀组与破血药组组间比较,差异无显著性意义(P>0.05);破血药组Bcl-2表达高于活血药组(P<0.05)。空白组、他汀组、活血药组、破血药组Bax表达明显低于模型组(P<0.01,P<0.05),空白组、他汀组、破血药组组间比较,差异无显著性意义(P>0.05),破血药组Bax表达低于活血药组(P<0.05)。空白组、他汀组、破血药组凋亡率明显低于模型组(P<0.01);破血药组凋亡率低于活血药组(P<0.05)。结论:破血药三棱、莪术通过上调Bcl-2的表达,下调Bax的表达,起到抑制细胞凋亡的作用,此作用优于活血药当归、川芎。Objective: To discuss the influence of blood-activating herbs of Radix Angelicae Sinensis and Rhizoma Chuanxiong as well as stasis-eliminating herbs of Rhizoma Sparganii and Rhizoma Curcumaeon the B cell leukemi- a-lymphoma-2(Bcl-2) and Bcr-2-associated X proteingene(Bax) protein expression and cell apoptosis in rat models of atherosclerosis(AS). Methods, Sixty male Wistar rats were equally randomized into 5 groups, blank control group, model group, blood-activating herbs group, stasis-eliminating herbs group and statins group. AS rat models were induced by high fat feeding. After establishing the model, the rats were given intragastric administration of the corresponding drugs according to the experimental design for 4 weeks. At the end of the experiment, the Bcl-2 and Bax Protein expression was examined and the cellular apoptosis rate was detected. Results, Bcl-2 expression was higher in stasis-eliminating herbs group and statins group than that in the model group(P 〈 0.01), but the difference between stasis-eliminating herbs group and statins group was insignificant. Stasis-eliminating herbs group had higher Bcl-2 expression level than statins group(P 〈 0.05). Bax expression was lower in the blank control group, stasis-eliminating herbs group blood-acti- vating herbs group and statins group than that in the model group(P 〈 0.01), but the difference among the former three groups was insignificant. Stasis-eliminating herbs group had higher Bax expression level than blood-activating herbs group(P〈 0.05). The apoptotic rate in the blank control group, stasis-eliminating herbs group and statins group than that in the model group(P 〈 0.01), and stasis-eliminating herbs group had lower apoptotic rate than blood-activating herbs group(P 〈 0.05). Conclusion: Rhizoma Sparganii and Rhizoma Curcumaeon have better effect than Radix Angelicae Sinen- sis and Rhizoma Chuanxiong on inhibiting apoptosis by up-regulating Bcl-2 expression and by down-regulating Bax ex- pression.
