摘 要:目的建立地榆药材总皂苷及地榆皂苷-I含量的测定方法,并对不同批次地榆药材中总皂苷和地榆皂苷-I的含量进行测定。方法采用优化条件的紫外-可见分光光度法建立了地榆药材中总皂苷的含量测定方法,采用HPLC—ELSD法测定地榆药材中地榆皂苷-I的含量。结果优选出总皂苷含量测定的显色体系为香草醛-冰醋酸-高氯酸,反应条件为15%香草醛,1.0mL高氯酸,80℃反应15min,经测定,地榆药材中总皂苷含量为7.25%~8.41%;HPLC—ELSD法9n4得地榆药材中地榆皂苷-I的含量为5.21%~6.18%。结论香草醛-冰醋酸-高氯酸法以及HPLC—ELSD法快速、简便、准确、重现性良好,分别适用于测定地榆药材中总皂苷和地榆皂苷-I的含量。Objective To establish the quantification method for total saponins and ziyuglycoside I, and to determine their contents in different batches of Sanguisorba officinctlis L. Methods The ultraviolet-visible spectrophotometer was optimized for the determination of total saponins, and HPLC-ELSD was employed for the determination of ziyuglycoside I in S. officinolis, respectively. Results The coloration system for the determination of total saponins was vanillin- acetic acid-perchloric acid. The condition of coloration was 15 % vanillin, 1 mL perchloric acid, 80 ~C water bathed for 15 min, and the detection wavelength was 549 nm. In seven batches of commercial crude drugs of S. officinolis, the contents of total saponins were measured as 7.25%-8.41%, and the contents of Ziyu-glycoside I were 5.21%- 6.18 %. Conclusion The developed methods are simple, rapid, accurate, stable, and reproducible with high selectivity, and are suitable for the determination of total saponins and ziyuglycoside I in the quality control of S. officinalis, respectively.
