摘  要:目的研究脂多糖(LPS)对健康人皮肤成纤维细胞增殖的影响,以阐明LPS在皮肤创伤愈合中的可能作用。方法体外培养健康人皮肤成纤维细胞,通过四甲基偶氮唑蓝(MTT)比色分析法、细胞计数、流式细胞术观察不同浓度的LPS(0、0.005、0.010、0.050、0.100、0.500和1.000μg/mL)对成纤维细胞增殖的影响。结果 LPS刺激浓度在0.005μg/mL时,成纤维细胞增殖率增高;随着刺激浓度增加,刺激作用增强;但当LPS浓度为0.5μg/mL时,促增殖作用开始降低;当浓度达到1.0μg/mL时则呈现抑制效应。结论在一定浓度范围内LPS促进成纤维细胞增殖,表明LPS在皮肤创伤愈合中可能具有重要作用。Objective To investigate the influence of lipopolysaccharide (LPS) on the proliferation of nor- mal human skin fibroblasts and its relation with wound healing. Methods Normal skin fibroblasts were exposed to LPS for a certain period of time. Then proliferation of fibroblasts was determined with the colormetric thiazolyl- blue (MTT) assay and cell counting. Results The proliferation of normal skin fibroblasts increased when chal- lenged with LPS at the concentration of 0. 005 μg/mL, and the increase showed a concentration dependent man- ner. However, when the concentration of LPS was 0.5 μg/mL,the influence of LPS on the proliferation of normal skin fibroblasts began to decrease; When the concentration of LPS reached 1.0 μg/mL,the proliferation of fibro- blasts was inhibited. Conclusion LPS could promote the proliferation of normal human skin fibroblasts within a certain extent,suggesting that LPS might play an important role in wound healing.
