摘  要:目的探讨磷酸酶基因(phosphataseandtensin homology deleted on chromosome ten,PTEN)在宫颈癌组织中的表达及其与宫颈癌临床生物学行为的关系。方法选取山东省潍坊市益都中心医院妇科2009年10月至2011年8月行门诊活检及手术切除的病理标本,其中宫颈癌组织59例、宫颈上皮内瘤变(CIN)组织21例及正常宫颈组织20例。采用免疫组织化学方法(SP法)检测各组织中PTEN表达情况,分析其表达情况与宫颈癌患者性别、年龄、分化程度、浸润深度、淋巴结转移、临床分期等临床生物学行为的关系。结果宫颈癌组织中PTEN的阳性表达率为38.98%(23/59),低于正常宫颈黏膜中的阳性表达率100.0%(20/20),二者比较差异有统计学意义(P<0.05)。宫颈癌组织中PTEN的表达随着癌灶的增大、分化程度的降低、浸润深度的加深、淋巴结的转移、临床分期的增加而明显降低。结论 (1)PTEN基因的失活参与了宫颈癌的发生。(2)PTEN的低表达预示着随着宫颈癌的进展和分化程度的降低,其侵袭和转移能力逐渐增强。Objective We aim to determine the expression of phosphataseand tensin homology deleted on chromosome ten (PTEN) in cervical cancer and to study the relationship of their expression with clinicpathological characteristics and to demonstrate the potential use of them as a prognostic and therapeutic agent. So that to look for the action of the marker in invasion and metastasis especially. Methods Total 59 specimen between Oct. ,2009 and Aug. ,2011 at our Hospital. Im- munohistochemical streptavidin-biotin-protein-linked peroxidase (SP) method was used to detect the expression of PTEN in 21 cases of CIN tissue, 20 cases of normal cervical tissue,59 cases of cervical cancer. It was analyzed that the PTEN expression and clinical stage, histological grade and other clinicopathological parameters of relevance. Results PTEN ex- pression in cervical cancer tissues (38.98%)were significantly lower than those in para-cancerous normal cervical tissue ( 100. 0% ) (P 〈 0. 05 ). The expression of PTEN was correlated with different stage and lymph node metastasis, and the expression of PTEN was also correlated with histological grade. Conclusions ( 1 ) The expression of PTEN in cervical cancer was higher than that in normal cervical tissues and CIN. (2) It suggested that expression of PTEN may play a role in the carcinogenesis of cervical cancer.
