摘 要:目的观察阿奇霉素、乙酰螺旋霉素、青蒿琥酯及联合用药对小鼠体内弓形虫的作用效果,探索治疗弓形虫病的有效药物及方法。方法小鼠腹腔接种RH株弓形虫速殖子5×104个/ml,建立急性弓形虫感染动物模型。设立阿奇霉素、乙酰螺旋霉素、青蒿琥酯单药治疗组以及阿奇霉素和乙酰螺旋霉素、阿奇霉素和青蒿琥酯、青蒿琥酯和乙酰螺旋霉素联合治疗组,分别于感染后2h给药,首次剂量加倍,疗程7d,以小鼠腹腔灌洗液虫体形态、小鼠生存时间和存活率作为观察指标,连续观察30d,评价单药和联合用药的治疗效果。实验设不治疗对照组。结果各治疗组小鼠体内的弓形虫速殖子形态发生改变,小鼠平均存活时间较对照组延长(P<0.05),以阿奇霉素单药、阿奇霉素和青蒿琥酯联合用药、青蒿琥酯单药治疗组最为显著,分别为(10.63±0.75)d、(7.37±0.36)d、(6.48±0.25)d,未治疗对照组为(4.33±0.22)d。结论 3种药物均有抗小鼠体内弓形虫作用,阿奇霉素、青蒿琥酯及二者联合用药作用效果更强。Objectives To observe the effect of azithromycin, acetyl spiramycin, and artesunate individually and in com-bination to counter toxoplasmosis and to explore effective drugs and therapies to treat toxoplasmosis. Methods Mice were inoculated intraperitoneally with 5 × 10^4 tachyzoites/ml of the RH strain of Toxoplasrna gondii in an animal model of acute T. gondii infection. One group received azithromycin alone, one received acetyl spiramyein alone, one received ar-tesunate alone, one received a combination of azithromycin and acetyl spiramycin, one received a combination of azithro-mycin and artesunate, and one received a combination of artesunate and acetyl spiramycin. Two h post-infection, the ini- tial dose was doubled. After 7 d of treatment, the morphology of parasites in peritoneal lavage fluid and the survival time and survival rate of mice were noted as indicators. Observation continued for 30 days. Results The morphology of Tox-oplasma tachyzoites changed significantly in treated groups, and treated groups had a longer average survival than the con- trol group (P(0.05). Specifically, survival for the group treated with azithromycin alone was 10.63 ± 0.75 d, that for the group treated with a combination of azithromycin and artesunate was 7.37 ± 0.36 d, and that for the group treated with artesunate alone was 6.48 ± 0.25 d. Survival for the untreated control group was 4.33 4± 0.22 d. Conclusion Three drugs had a potent effect on Toxoplasma in mice. Azithromycin, artesunate, and combinations of the two were more potent.
