摘  要:研究了镰刀霉菌(Fusarium sp)JN158色素的提取条件、溶解特性和液相分离方法,实验结果表明:该紫色素的最佳提取溶剂为纯乙酸;色素只能溶解于酸性的有机溶剂或有机酸中;色素的最佳液相分离条件是:色谱柱,Phecda C18(5μm,4.6 mm×250 mm);柱温:25℃;DAD检测器检测波长:254 nm;流量:1.0 mL/min,进样量:10μL,流动相A,0.1%TFA的乙腈(V:V),流动相B,0.1%TFA的蒸馏水(V∶V),梯度洗脱条件:流动相A,0-3 min,2%增到6%,3-3.1 min,6%增到45%,3.1~28 min,45%增到70%,28-32 min,70%增到80%;分离得到8种色素,在酸性乙腈中,1种呈现黄色,4种呈现红色,3种呈现橙色,按出峰顺序,8种色素的吸收峰分别为508、458、456、498、510、456、454、506 nm。The conditions of extraction and separation and the dissolution characteristics on the pigment of fusarium are studied. The results of Experimental show that the best extraction solvent of the pigment is pure acetic. The pigment only can dissolve in organic acids and acidic organic solvents. The best high-performance liquid separation conditions of the pigment are the followings: the column is Phecda C18 (4.6 min×250 mm,5 mm). The temperature of the column is 25℃. The detection wavelength of the DAD detector is 254 nm. The flow rate is 1.0 mL/min. The injection volume is 1 μL. The mobile phase A is acetonitrile containing one-thousandth of trifluoroacetic acid. The mobile phase B is distilled water containing one-thousandth of trifluoroacetic acid. The gradient elution program is thai : mobile phase A, 0 N3 rain, 2%-6% ; 3 -3.1 rain, 6%-45% ; 3.1-28 rain,45%-70% ;28-32 rain,70%-80%. Eighl kinds of pigmenls are obtained,and four kinds of pigmenls are red,three orange,one yellow in acidic acetonitrile. In accordance with the order of the peaks, their maximum absorplion peaks are 508,458,456,498,510,456,454,506 nm respectively.
