Experimental and Clinical Study on Circadian Rhythms of Melatonin and the Concentration of Free Calcium in Alzheimer' s Disease

摘  要:目的:通过动物实验结合临床研究阿尔茨海默病褪黑素与游离钙离子浓度昼夜变化,探讨褪黑素抵抗神经细胞对钙离子吸收的调控机制,为临床防治阿尔茨海默病提供新的思路。方法:采用ELISA法定量测定昼夜不同时间点正常小鼠与阿尔茨海默病模型小鼠血清及脑浆的褪黑素电极法测定游离钙离子浓度;并同步测定正常老人与阿尔茨海默病老人血清褪黑素、游离钙离子浓度,用余弦函数分析昼夜节律。结果:正常小鼠和正常老人褪黑素分泌存在昼夜节律,阿尔茨海默病的小鼠和老人夜晚褪黑素分泌减少昼夜节律消失。结论:褪黑素的降低与神经组织噬钙离子增大可能是阿尔茨海默病的危险因素。OBJECTIVE : To explove melatonin resistance of nerve cells to calcium absorption regulation mechanism by observing the diurnal variation of melatonin and free calcium concentration in Alzheimer' s disease (AD) in order to provide a new train for the clinical prevention and treatment of Alzheimer' s disease. METHODS: Circadian at different time points in normal mice and mice model of Alzheimer' s disease was determined by using ELISA method and free calcium ion concentration was also determined by brain serum melatonin electrode method. The concentration of serum melatonin, free calcium ion were tested in normal elderly and the elderly with Alzheimer disease by using the cosine function analysis of circadian rhythm. RESULTS: The circadian rhythm of melatonin secretion exited in normal mice and elderly. But night melatonin secretion in the elderly and the mice with AD reduced circadian rhythm. CONCLUSION : The reduces of melatonin and the increases of nervous tissue macrophage calcium may be the risk factors of AD.
