摘  要:目的探讨硫酸软骨素(CS)对Lewis肺癌的抑制作用及其分子作用机制。方法用MTT法检测CS对小鼠Lewis肺癌细胞增殖的影响。建立小鼠Lewis肺癌模型,分为模型对照组(生理盐水0.2 mL/d)、CS高[500 mg/(kg.d)]、低[100 mg/(kg.d)]剂量组和环磷酰胺组[50 mg/(kg.w)],分别腹腔注射给药。记录各组小鼠的体重、生存时间和生活状况,计算脾指数、胸腺指数和抑瘤率。结果随着CS浓度增大,小鼠Lewis肺癌细胞的增殖能力明显降低。CS各剂量组小鼠精神状态、身体状况较模型对照组和环磷酰胺组好,CS能明显提高胸腺指数、脾指数和抑瘤率,高剂量组各指标高于低剂量组。结论 CS能明显减弱小鼠Lewis肺癌细胞的增殖能力,提高Lewis荷瘤小鼠生活质量,并对肿瘤生长有显著地抑制作用,此结果可能是通过抑制癌细胞增殖、提高小鼠免疫力得到的。Objective To investigate the anti-cancer effect of chondroitin sulfate and explore its anti-tumor mechanism. Methods The capability of proliferation of Lewis lung carcinoma cells were individually determined by MTT. The mouse Lewis pulmonary carcinoma models were establish and divided randomly into 4 groups: model group, chondroitin sulfate high dose group, 500 mg/(kg·d), chondroitin sulfate low dose group, 100 mg/(kg·d) and cyclophosphamide group, 50 mg/(kg·w). The medicine was given separately by intraperitoneal injection. Record the changes in body weight and mental state, and calculate the thymus index, spleen index and inhibition rate. Results The proliferations of the Lewis lung carcinoma cells were reduced with increasing dose in the chondroitin sulfate groups compared with the control group. The mental and physical state of the mice in chondroitin sulfate groups was better than those in other groups. Chondroitin sulfate improved the thymus index, spleen index significantly. Conclusion Chondroitin sulfate can reduce the proliferations of the Lewis lung carcinoma cells, improve the living quality of mice which have Lewis lung cancer, and inhibit the tumor growth significantly. The effect may be achieved by killing the cancer cells and inhibiting the angiogenesis.
