产益康栓对大鼠阴道黏膜刺激及过敏反应的实验研究

摘　　要：目的:观察产益康栓对大鼠阴道黏膜的刺激及过敏反应,为临床安全用药提供理论依据。方法:①阴道黏膜刺激试验:将16只大鼠随机分成单次涂抹组、多次涂抹组,每组8只。各组给药剂量为0.4 g/(kg.d)。单次涂抹组:涂抹后24小时用温水洗去残留的药物。多次涂抹组:连续8天,最后1次涂抹24小时后用温水洗去残留的药物,并分别计算阴道黏膜刺激性反应平均积分值。②大鼠阴道过敏试验:将24只大鼠分为产益康栓高、低剂量组,对照组,每组8只。产益康栓高、低剂量组分别按0.8、0.4 g/(kg.d)给药,隔日1次,共5次。对照组给予同体积0.9%生理盐水注射液,隔日1次,共5次。给药10天后观察各组大鼠阴道黏膜变化情况。结果:观察期间,单次涂抹组、多次涂抹组活动均正常,无死亡现象,2组阴道黏膜刺激性反应平均积分值为0分和0.17分。产益康栓高、低剂量组,对照组大鼠阴道无充血、水肿等症状,致敏率为0.0%。结论:产益康栓对大鼠阴道黏膜组织无明显的刺激性,未发现明显的过敏反应。Objective： To provide theoretical evidence for safe medication through observing the effects of ChanYiKang suppository on the irritation and allergy of vaginal mucosa in rats. Method：①Stimulation test of vaginal mucosa： all 16 rats were randomized into the groups of single smearing and smearing for many times, eight rats each group. The dosage given to each group was 0.4 g/ （kg·d）. The groups of single smearing： the residual medicine was washed away with warm water in 24 hours after smearing. The groups of smearing for many times： for eight consecutive days, till the last time the residual medicine was washed away with warm water in 24 hours. Average score of irritative response in vaginal mucosa was calculated respectively. ②Vaginal allergic tests of rats： all 24 rats were separated into high and low dose groups of ChanYiKang suppository, blank control group, eight rats each group. High and low dose groups of ChanYiKang suppository were medicated in the dosage of 0.8 g/（kg·d）and 0.4 g/（kg ·d）, once every other day, for five times. Blank control group received the same volume of 0.9% physiological saline, once every other day, for five times. Changes of vaginal mucosa in all groups were observed in ten days after medi- cating. Result： During the period of observation, the rats in both groups were normal and there was no death. Aver- age scores of irritative response in vaginal mucosa were zero and 0.17. There was no congestion, edema and others happened in high and low dose groups of ChanYiKang suppository as well as blank control group. Sensitization rate was 0.0%. Conclusion： ChanYiKang suppository demonstrates neither obvious irritation nor allgery to vaginal mu- cosa of rats.
