摘  要:为了解中国地区宫颈癌病人中人乳头瘤病毒16 型E6E7 基因结构特点,从中国山东地区宫颈癌活检组织中提取组织DNA,经HPV 多重引物PCR 法鉴定标本中感染HPV 型别,选单纯感染HPV16 型两例标本DNA为模板进行PCR 扩增,获得HPV16 E6E7 基因后,重组入pALTER-1 载体,进行双向测序、分析。DNA 序列分析表明:两例标本的HPV16 E6E7 序列全长均为776bp, 与已发表的德国标准株长度相等,两例均为变异株,共检出5 处核苷酸变异位点,其中4 处可导致氨基酸改变。中国山东地区宫颈癌病人中HPV16 E6E7 的基因结构与德国标准株的HPV16 E6E7 基因之间存在差异。To study the structure specificity of HPV16E6E7 gene of Chinese cervical carcinoma patients in Shandong province The tissue DNA were extracted from cervical carcinoma biopsies of Shandong province and then the types of HPVs which infected in cervical carcinoma tissues were identified by HPV multiple primers PCR HPV16E6E7 gene was amplified by PCR from the cervical carcinoma tissue DNA with the infection of HPV16 type only and then cloned the E6E7 gene into pALTER-I vector After sequencing the double strand the gene was compared with the prototype E6E7 gene of HPV16 Sequencing results indicated there were sequence diversity in two cases and total of five nucleotide exchanges were detected four of these led to amino acid exchanges there are structure differences between the HPV16E6E7 of Chinese and that of the standard strain
