摘  要:目的:观察阿托伐他汀对动脉粥样硬化(AS)早期病变的作用。方法:雄性新西兰白兔随机分为正常对照组(正常饮食)、动脉粥样硬化组(高脂饮食)和阿托伐他汀组,10周后测定血清中血脂、肿瘤坏死因子-α(TNF-α)、C反应蛋白(CRP)及白介素-10(IL-10)水平,H-E染色测定胸主动脉内膜及中膜厚度,免疫组织化学法测定胸主动脉环氧化酶2(COX-2)、氧化低密度脂蛋白受体1(LOX-1)及α肌动蛋白(α-actin)的表达。结果:高脂饮食后兔血清总胆固醇(TC)、甘油三酯(TG),低密度脂蛋白胆固醇(ox-LDL)浓度升高,胸主动脉形成典型脂质条纹。与动脉粥样硬化组相比,阿托伐他汀组血清血脂、ox-LDL、 TNF-α、 CRP水平降低,IL-10水平则增高。胸主动脉内膜厚度和内膜/中膜厚度比下降,主动脉COX-2、 LOX-1、 α-actin的表达降低。结论:阿托伐他汀通过抑制血管炎症反应及平滑肌细胞增殖、降脂等多环节抑制动脉粥样硬化斑块的早期形成。Objective: To observe the effect ofatorvastatin on the progression of atherosclerosis (AS). Methods: The New Zealand male rabbits were divided randomly into 3 groups (n= 7), including normal control group (normal diet), AS group (high-cholesterol diet), atouast group. All rabbits were sacrificed after 10 weeks. Blood was drawn for quantify the serum lipid levels and the levers of creactive protein (CRP), tumor necrosis factor-α(TNF-α) and interleukin-10 (IL-10). Thoracic aorta were obtained for hematoxylin-eosin (HE) staining and cycloxygenase; epoxidase-2 (COX-2), lectin-like oxidized low density lipoprotein-1 (LOX-1) and α-acttn immmunohistochemistry assay. Results: In all cholesterol-fed rabbits, the level of serum TC, TG, LDL and α-LDL were increased remarkably. The typical fatty streaks on the aorta had been formed. Com- pared to AS group, atorvastatin could decrease serum lipid, atorvastatin also could decrease serum CRP, TNF-α level and in- crease serum IL-10 level significantly. Atorvastatin can downregulate the COX-2, LOX-1 and α-actin expression significantly as compared with AS group. Conclusion: Atorvastatin can suppress vascular inflammation, inhabit.smooth muscle proliferation and lower blood lipid, all these contributed to inhibit the formation of the early atheroselerotic lesion in cholesteral-fed rabbits.
