摘 要:目的:探讨不同剂量电子射线局部照射对小鼠下颌下腺的放射损伤效应。方法:将60只雌性C57BL/6小鼠分为6组(n=10),用直线加速器对每组小鼠分别给予0(对照)、12、15、18、21、24 Gy进行头颈部局部电子射线照射,照射后1月观察小鼠体重变化,照射后1周、1个月进行血象分析,照射后2、3、4月进行唾液流量测定,切片HE染色,透射电镜观察以及细胞凋亡检测。结果:照射组小鼠在照射后体重轻微下降,血常规各项较对照组下降,唾液流量率较对照组有明显下降,而且随时间延长,照射剂量越大,降低幅度也越大。HE切片显示照射组下颌下腺组织明显损伤,腺泡细胞数量减少,结构紊乱,剂量越大,损伤越明显;电镜下见腺泡细胞水肿变性坏死,细胞器结构紊乱;细胞凋亡指数为(21±2.5)%;18 Gy照射后2月炎性细胞浸润明显,唾液流量降低约60%。结论:随着照射剂量的增加下颌下腺损伤越严重,18 Gy电子射线照射小鼠涎腺引起的的损伤是较理想的涎腺放射损伤模型。Objective: To study the changes in submandibular glands of mice after electron rat irradiation with different doses. Methods: 60 female C57BL/6 mice were divided into 6 groups(n = 10). The submandibular glands of mice were locally electron rat irradiated(IR) with a single dose of 0(the control), 12, 15, 18, 21 and 24 Gy respectively via a liner accelerator. Weight of the mice was measured 1 month after IR. Blood examination was performed 1 and 4 weeks after IR. Saliva flow rate was measured 8, 12 and 16 weeks after IR. The morphologic changes were observed by HE staining and TEM examination. The cell appoptosis was tested by TUNEL kit. Results: The body weight was slightly decreased in irradiated mice after IR. Blood routine measurements were de- creased, saliva flow rate was decreased in a IR dose- dependant manner. HE staining showed that the damage severity of the gland tis- sue was related to the IR dose, 2 months after IR at 18 Gy saliva rate deacreased by 60%, inflammatory infiltration was obvious. Aci- nar cell edema and degeneration necrosis were observered by TEM. the apoptosis index was (21 ± 2.5 )% in 18 Gy irradiated mice 3 months after IR. Conclusion: X-irradiation may dose-dependantly damage salivary gland. 18 Gy electron irradiation is recomended for the establishment of mouse submandibular gland irradiation damage model.
