摘  要:背景:目前发现退变椎间盘中均有细胞因子和炎症递质等物质存在,但他们是来源于椎间盘外,还是椎间盘细胞自我产生没有定论。目的:检测各种腰椎间盘细胞能否自分泌产生白细胞介素1β。方法:分离培养兔腰椎间盘的各种细胞,对培养的细胞进行鉴定,用Fn120的纤维粘连蛋白碎片进行细胞诱导,以PBS作为对照。用RT-PCR方法和免疫荧光方法检测各种细胞内白细胞介素1β的表达情况。结果与结论:经Fn120的纤维粘连蛋白碎片诱导的实验组椎间盘软骨细胞、纤维环细胞均能产生白细胞介素1β。结果表明椎间盘软骨终板细胞、纤维环细胞均能自分泌产生白细胞介素1β。BACKGROUND: The studies found that there are cytokines and inflammatory cytokines in the degenerative intervertebral disc, but it is still unknown that whether they come from the outer intervertebral disc or inner intervertebral disc. OBJECTIVE: To investigate whether the cells of different lumbar discs can produce inflammatory cytokine interleukin-1β.METHODS: The cells of rabbit lumbar disc were isolated and cultured and then identified. The cells were induced with Fn120 fibronectin fragments, and PBS was used in the control group. The expression of interleukin-1β was detected by immunofluorescence method and reverse transcription-PCR,RESULTS AND CONCLUSION: In the experimental group, the cartilage cells of intervertebral disc and fibroblasts of annulus fibrosis could produce the interleukin-lβ. The results showed that cartilage endplate cells and the fibroblasts of annulus fibrosis can produce interleukin-lβ.
