摘  要:目的观察护卵汤对丙氨瑞林(GnRHa)超排卵大鼠卵巢细胞因子及受体的影响,为护卵汤辅治辅助生殖技术(ART)提供理论依据。方法模仿人类辅助生殖中的长周期,对大鼠进行GnRHa垂体降调节预处理后再用HMG等药物超排卵(模型组),同时给予中药护卵汤辅助治疗(护卵汤组),以及自然周期未使用任何药物的大鼠为正常对照(正常组)。在注射绒毛膜促性腺激素(HCG)日摘取卵巢组织用抗体芯片法检测卵巢细胞因子及受体的表达。结果与正常组相比,模型组卵巢微环境中生长激素(GH)、生长激素受体(GHR)、白细胞介素-1β(IL-1β)等十几种细胞因子及受体表达减少,瘦素(Leptin,OB)、γ干扰素(IFN-γ)等表达增多;与模型组相比,护卵汤组卵巢微环境中GHR、IL-1β、转化生长因子-β1(TGF-β1)等十几种细胞因子及受体表达增多,IFN-γ、Leptin(OB)、肿瘤坏死因子(TNF-α)等表达减少。结论 GnRHa超排卵对大鼠卵巢微环境有不利影响;护卵汤能改善GnRHa超排卵大鼠卵巢微环境,从而有利于卵泡发育及卵子质量。Objective To observe the effects of Huluantang upon contents of cytokines in ovary in rats under GnRHa controlled ovarian hyperstimulation(COH), and to provide related basis for the effectiveness of Huluantang in assisted reproductive technology(ART). Methods After receiving GnRHa pituitary down regulation pretreatment which imitates the human long period of COH, a number of rats were separated into a so-called model group treated with HMG/ HCG superovulation drugs and so-called Huluantang group treated with HMG/HCG superovulation drugs and Chinese medicine of Huluantang. The natural cycle of the rats were compared to a normal group receiving no medicine. The contents of cytokines in ovary were observed by antibody array on the day of HCG injection. Results Compared with those in the normal group,the levels of some cytokines such as GH,GHR,IL-1β in ovary were decreased above 1.5 times, the levels of Leptin (OB), IFN-γ, Fas/TNFRSF6, TRAIL in ovary were increased above 1.5 times; Compared with those in the model group, the levels of some cytokines such as GHR,IL-1β ,TGF- β1 in ovary were increased above 1.5 times, the levels of Fas/TNFRSF6, IFN- γ, Leptin (OB), TNF-α in ovary were decreased above 1.5 times in the Huluantang group. Conclusion The GnRHa controlled ovarian hyperstimulation may have adverse effects on the ovarian microenvironment in rats; Huluantang can improve the ovarian microenvironment, which can promote the follicular development and the quality of occytes in rats under GnRHa controlled ovarian hyperstimulation.
