芩蒿滴鼻剂对变应性鼻炎大鼠疗效的实验研究

摘　　要：目的考察芩蒿滴鼻剂对变应性鼻炎大鼠模型的治疗作用。方法 50只SD大鼠,以0.1%卵清蛋白(OVA)和氢氧化铝佐剂混悬液腹腔注射进行全身致敏,用2%卵清蛋白溶液进行鼻部抗原攻击,建立变应性鼻炎大鼠模型。将造模成功大鼠随机分为模型组、滴通鼻炎水组、芩蒿滴鼻剂低剂量组[0.1g浸膏(相当于生药量2.12g)/mL]、芩蒿滴鼻剂高剂量组[0.5g浸膏(相当于生药量10.58g)/mL)]4组,并设正常对照组。将芩蒿滴鼻剂滴鼻给药6d后,用酶联免疫吸附(ELISA法)测定血清卵清蛋白特异性IgE(OVA-sIgE)含量,取鼻黏膜做病理组织学检查。结果模型组血清OVA-sIgE含量明显高于正常对照组,差异有统计学意义(P<0.01);芩蒿滴鼻剂高、低剂量组血清OVA-sIgE含量低于模型组,差异有统计学意义(P<0.01);芩蒿滴鼻剂高、低剂量组之间差异无统计学意义。病理学结果显示,模型组鼻黏膜部分黏膜上皮坏死、脱落,固有层血管不同程度的扩张、充血,伴有大量嗜酸粒细胞及其他炎性细胞的浸润;芩蒿滴鼻剂高、低剂量可明显减轻搔鼻、打喷嚏、流鼻涕及鼻塞等典型的鼻部过敏症状,改善鼻黏膜毛细血管的扩张、充血,抑制嗜酸性粒细胞浸润。结论芩蒿滴鼻剂能降低变应性鼻炎大鼠血清OVA-sIgE含量并改善鼻黏膜病理改变,对变应性鼻炎模型大鼠具有一定的治疗作用。Objective To study the therapeutic effect of Qin-hao nose drops on allergic rhinitis in rats. Methods Fifty healthy rats were sensitized by intraperitoneally injecting the suspension of 0.1% ovalbu min （OVA） and AI（OH）3, and intranasally challenged with 2O/oo OVA to establish allergic rhinitis models. The successful model rats were randomly divided into group, Qin-hao nose drops little-dosage group [-0. 1 four groups, the model group, Di-tong nose drops g extract （equivalent to the amount of crude drug 2.12 g）/mL], Qin-hao nose drops high-dosage group [-0.5 g extract （equivalent to the amount of crude drug10.58 g）/mL] and the normal control group. After 6 days, the level of serum sIgE was determined by ELISA and observed the nasal mucosa by light microscope. Results The serum OVA-Immunoglobulin E （sIgE） of the model group were significantly higher than the normal control group （P %0.01） ; the serum OVA-sIgE of Qin-hao nose drops high, little-dosage group were lower than the models group （P %0.01） ; the difference of between Qin-hao nose drops high-dosage and little-dosage group had no statistically signif icant . Pathology results showed that in the morphology nasal mucosa in allergic rhinitis model , there were breaking off, necrosis, blood vessels of different degrees of expansion, congestion, accompanied by a large number of eosinophils and mast cells infiltration; Qin-hao nose drops high-dosage and little-dosage group can obviously reduced scratching nose and sneezing, runny nose and stuffy nose and other typical of the nasal allergy symptoms, an ophils infiltration. Conclusion d impr Qin-h ove the nasal mucosa capillary expansion, congestion, restrain eosin- ao nose drops can reduce the serum OVA-sIgE content in allergic nitis rats and improve the nasal mucosa pathological change. Qin-hao nose drops have certain effective treatment on allergic rhinitis in rats
