地榆总皂苷对小鼠腹水型H22肝癌的抑制作用及机制研究  被引量:8

摘  要:目的:探讨地榆总皂苷体内抗肿瘤的作用及其机理。方法:通过腹腔接种H22肝癌细胞建立荷瘤小鼠模型,并按体质量随机分为阴性对照组(0.5%CMC溶液)、阳性对照组(呋喃氟尿嘧啶,120mg/kg)、地榆总皂苷大(4g/kg)、中(2g/kg)、小(1g/kg)剂量组,连续灌胃给药10天后,观察记录60天内动物死亡时间,计算小鼠生存天数及中位生存时间(MST)。再通过上述方法建立荷瘤小鼠模型,用地榆总皂苷灌胃给药,连续10天后,抽取小鼠腹水,采用荧光染色法及流式细胞术检测细胞凋亡。结果:各组MST分别为14.51、20.2、19.2、21.0、17.5天,阳性对照及各给药组与阴性对照组MST比值分别为139.3%、132.4%、144.8%、120.7%。荧光显微镜下观察发现阴性对照组细胞核呈蓝色,给药组细胞形态改变,细胞核呈致密浓染,有的细胞核呈碎块状致密浓染,颜色发白。地榆总皂苷大、中、小剂量组细胞凋亡率分别为1.05%、1.14%、0.79%,较阴性对照组明显增高(P<0.05)。各给药组细胞发生不同程度凋亡。结论:地榆总皂苷具有明显的体内抗肿瘤活性,其抗肿瘤活性机理为诱导肿瘤细胞发生凋亡。Objective: To study the antitumor effect and mechanism of Sanguisorba saponins in vivo.Methods:Mice H22-ascites tumor models were established and divided randomly into 5 groups as follows: negative control group(0.5% CMC),positive control group(FT-207,120mg/kg),Sanguisorba saponins high dose group(4g/kg),medium dose group(2g/kg),low dose group(1g/kg).After intragastric administrated 10 days continuously,death time of each mouse was recorded during 60 days,then survival day and median survival time(MST)were calculated.Once more,after intragastric administrated 10 days continuously,ascites of each mouse was collected,fluorescein staining and flow cytometry were used to test cell apoptosis.Results: MST of each group was 14.51,20.2,19.2,21.0,17.5 days respectively,the ratio of MST of each administrated group to negative control was 139.3%,132.4%,144.8%,120.7% respectively.Cells of each administrated group demonstrated morphous change,compact and thick of the nucleolus under fluorescent microscope,the ratio of apoptosis was 1.05%,1.14%,0.79% respectively.Apoptosis occured in ascites cells of each administrated group.Conclusion: Sanguisorba saponins have a significantly antitumor effect in vivo and the mechanism is to induce ascites cells to apoptosis.
