猪流行性腹泻病毒ShT株的分离及传代培养

摘　　要：从广东省某猪场疑为病毒性腹泻的发病猪采集腹泻粪便样品,以PCR方法扩增出猪流行性腹泻病毒N基因后,采用细胞培养法进行病毒分离。用套式PCR、胶体金试纸卡和血清中和试验对细胞分离物进行检验,证实其为一株猪流行性腹泻病毒,命名为ShT株。将猪流行性腹泻病毒ShT株在Vero细胞上进行传代培养,观察其培养特性。结果表明,PEDV ShT株能在Vero细胞稳定传代,第20代、25代、30代病毒液的TCID50/0.1 mL分别为10-7.0、10-7.13、10-7.33。A porcine epidemic diahorrea virus （PEDV） field strain was isolated from the feces of piglets suspected to be suffering from a severe diarrhea in Guangdong Province, and was identified by nested PCR, Anigen Rapid PEDV Ag test Kit and serological neutralization test, and named ShT. The strain was serially cells, and characteristics of the virus culture and virus titer were also explored. The TCID50/0.1 25th and 30th passage of PEDV ShT strain was respectively 10-7.0,10-7.13 and 10-7.33. passaged by vero mL of the 20th,
