摘  要:目的:观察纤维介素2(fgl2)基因沉默对心肌微血管内皮细胞(MMVECs)增殖、迁移及血管生成素-1、-2(Ang-1、Ang-2)mRNA表达的影响。方法:制作并包装人fgl2RNA干扰慢病毒fgl2-RNAi-LV。将细胞分为单纯MMVECs组(对照组)、GFP空载体慢病毒转染组(GFP组)和fgl2-RNAi-LV转染组。稳定转染4d后,应用实时定量RT-PCR法检测各组fgl2、Ang-1和Ang-2mRNA的表达,MTT法检测细胞增殖,Transwell法检测细胞迁移能力。结果:与对照组及GFP组比较,fgl2-RNAi-LV转染组fgl2mRNA表达明显下调(F=180.301,P<0.001),Ang-1、Ang-2mRNA的表达明显增加(F=59.450和54.572,P<0.001),MMVECs的增殖能力与迁移能力增强(F=5.341和6.122,P<0.05);对照组与GFP组比较差异无统计学意义(P>0.05)。结论:fgl2基因沉默促进MMVECs的增殖、迁移,其分子机制可能与Ang-1、Ang-2mRNA表达上调有关。Aim:To observe the effect of fibrinogen-like protein 2(fgl2) gene silencing on myocardial microvascular endothelial cells(MMVECs) proliferation and cell migration and whether Ang-1 and Ang-2 involve or not.Methods:We constructed a lentiviral RNA interferenced vector of human fgl2 gene.MMVECs were transfected by lentiviral vector.Four days after transfection,quantitative real-time RT-PCR was used to detect the expressions of fgl2,Ang-1 and Ang-2 mRNA,MTT assay was used to detect cell proliferation,and Transwell was used to detect cell migration.Results:The expression of fgl2 mRNA was decreased significantly compared with control group and GFP group(F=180.301,P0.001).While the expressions of Ang-1 and Ang-2 mRNA was increased(F=59.450 and 54.572,P0.001).The proliferation and migration ability of MMVECs increased(F=5.341 and 6.122,P0.05).Conclusion:Fgl2 gene silencing induces MMVECs proliferation and cell migration.The possible mechanism is that fgl2 gene silencing upregulates the expressions of Ang-1 and Ang-2 mRNA.
