摘  要:目的:研究磷酸肌酸钠(PCr)对缺氧窒息幼鼠肾组织缺氧诱导因子-1α(HIF-1α)和血管内皮生长因子(VEGF)表达的影响。方法:24只幼鼠随机分为PCr干预组(PCr组)、模型组和正常对照组,每组8只。PCr和模型组幼鼠制作缺氧窒息模型,PCr组幼鼠在制作模型前0.5h和缺氧窒息后即刻腹腔注射1.4mg/g的PCr。模型组在相同时间点腹腔注射等量的生理盐水。正常对照组不作处理。缺氧窒息后6h,切除各组幼鼠肾脏,采用HE染色法观察肾组织病理形态变化,RT-PCR和免疫组织化学法检测肾组织HIF-1α和VEGFmRNA和蛋白的表达。结果:模型组幼鼠肾组织可见肾小管上皮细胞肿胀,大量炎性细胞浸润,部分上皮细胞变性坏死,肾小球出现水肿、甚至固缩;PCr组幼鼠肾小管和肾小球受损状况明显减轻。3组幼鼠肾组织HIF-1α和VEGFmRNA及蛋白的表达比较,差异有统计学意义(F=196.805、484.734、90.410和79.329,P均<0.001),PCr和模型组均较正常对照组升高,PCr组较模型组降低(P<0.05)。结论:PCr可能通过下调HIF-1α、VEGF的表达减轻肾组织的缺氧性损伤。Aim:To research the influence of creatine phosphate sodium(PCr) on expressions of hypoxia-inducible factor-1 alpha(HIF-1α)and vascular endothelial growth factor(VEGF)in renal tissue of anoxia asphyxia young rats.Methods:A total of 24 young rats were randomly divided into three groups,PCr intervention group(PCr group),model group and normal control group,8 rats in each group.After anoxia asphyxia 6 h,shape and structure of the kidney were observed using HE staining,the expressions of HIF-1α and VEGF mRNA were detected using RT-PCR and the expressions of HIF-1α and VEGF protein were detected by immunohistochemical staining.Results:Compared with normal control group,pathological changes of renal lesions were observed in renal tissue of both PCr and model groups.The expressions of VEGF and HIF-1α mRNA and protein in renal tissue among the 3 groups had significant differences(F=196.805,484.734,90.410,and 79.329,P0.001),which were higher in both PCr and model groups than those in normal control group,and lower in PCr group than those in model group(P0.05).Conclusion:PCr could reduce kidney damage after anoxia asphyxia by down-regulating the expressions of HIT-1α and VEGF.
