摘 要:目的 确定ELISA实验检测乙型肝炎病毒表面抗原(HBsAg)的临界点浓度C50,从而得到乙型肝炎病毒表面抗原测定的"灰区"范围,并比较两种HBsAg检测试剂.方法 根据定性实验临界点的定义,采用酶联免疫吸附实验(ELISA),分别用两种HBsAg试剂测定一系列血清浓度的标本,确定HBsAg的临界点浓度C50和C5及C95浓度,继而分别确定两种HBsAg试剂的"灰区".结果 A试剂HBsAg C50浓度为0.40 IU/ml,"灰区"S/CO值为0.84~1.19;B试剂HBsAg C50浓度为0.35 IU/ml,"灰区"S/CO值为0.72~1.26.结论 HBsAg不同检测试剂的临界点浓度C50不一样,相应的"灰区"也不一样.实验室应对所用的试剂、方法进行评价,建立自己的C50浓度和"灰区"范围.Objective To get the gray-area of hepatitis B virus surface antigen (HBsAg) by determination C50 concentration HBsAg of ELISA,and compare with two kinds of testing reagents of HBsAg. Methods According to the definition of critical point of qualitative test,critical point concentration (C50) of HBsAg,C5 concentration and C95 concentration of HBsAg were determined with a series of different concentration samples using two kinds of HBsAg regents by enzyme linked immunosorbent assaysand gray-area of two kinds of HBsAg reagents was gained respectively. Results C50 concentration of A reagent was 0. 40 IU/ml and S/CO value of gray-area was 0. 84-1.19. C50 concentration of B reagent was 0.35 IU/ml and S/ CO value of gray-area was 0.72-1.26. Conclusion Serum concentration of C50 concentration of two kinds of HBsAg reagent was different,likewise the gray-area was different. Reagent and method should be evaluated in Laboratory. C50 concentration and gray-area of HBsAg should be set up.
