摘 要:目的探讨当归补血汤对大鼠局灶性脑缺血再灌注损伤的保护作用。方法雄性SD大鼠108只随机分为正常组、缺血再灌注组(模型组)和当归补血汤组(治疗组)。采用线栓法复制大鼠右侧大脑中动脉栓塞缺血再灌注模型。治疗组术后连续15 d以0.36 g/mL浓度当归补血汤3.6 g/kg灌胃,每日2次(早、晚各1次)。治疗10 d后进行各组大鼠神经行为学测试;TTC染色观察大脑皮质梗死体积变化;股静脉注入2%伊文斯蓝溶液观察血脑屏障通透性变化;Nissl染色法观察大鼠海马CA1区神经元的形态和数量。结果与缺血再灌注组比较,当归补血汤治疗组神经功能评分增高,大脑皮质脑梗死体积减少(P<0.05),伊文斯蓝的含量减少(P<0.05),神经元数量增加(P<0.05)。结论当归补血汤可改善脑缺血再灌注损伤大鼠的神经功能,减少脑梗死体积,减少大鼠海马CA1区神经元的死亡,对海马CA1区神经元具有保护作用。[Objective] To investigate the protective effect of the decoction of angelicae sinensis on focal cerebral ischemia-reperfusion injury with rats. [Methods] 108 male SD rats will be randomly divided into normal group, cerebral ischemia-reperfusion group (model group) and the decoction of angelicae sinensis-treat- ment group. A rat model of the right middle cerebral artery occlusion]reperfusion (MCAO) was created using filament method. Before being operated, treatment-group rats will be lavaged wish 0.36 g/mL Dong Quai Bu Xue Decoction 3.6 g/kg twice a day for 15 days (once in the morning and once in the evening). 10 days after operated, The defects of neurological behavior of the rats and the changes of cerebral infarct volume with TIC staining will be observed; Permeability changes of blood-brain barrier will also be observed after 2% Evans blue is injected into the femoral vein of the rats. The numbers and morphological change of neurons in the Hippocampal CA1 will be observed through Nissl staining. [Results] Compared with ischemia-reperfusion group, neurological function score increased in the decoction of angelicae sinensis- treatment group. TTC staining shows the infarct volume of Cerebral cortex in the decoction of angelicae sinensis were reduced (P 〈 0.05); The content of Evans blue in the Hippocampal CA1 was reduced (P〈0.05); The number of neurons of the the Hippocampal CA1 was increased (P〈0.05). [Conclusion] Behavioral capabilities of the focal cerebral ischemia-reperfusion injury neurological function with rats were improved by the decoction of angelicae sinensis, the death of neurons in the Hippocampal CA1 was alleviated, neurons of Hippocampal CA1 was protected.
