摘  要:目的研究尼莫地平(Nimodipine)对新生大鼠延髓脑片基本节律性呼吸放电的作用,为延髓呼吸中枢相关疾病的临床治疗和尼莫地平的使用提供思路和实验依据。方法制作新生大鼠离体延髓脑片标本,使用95%氧(O2)和5%二氧化碳(CO2)混合气饱和并持续通气的人工脑脊液(artificial cerebrospinal fluid,ACSF)灌流脑片,吸附电极吸附腹侧面舌下神经根,记录节律性呼吸放电(rhythmic respiratory discharge activity,RRDA)。稳定记录到RRDA后分别使用含0.000、0.025、0.050、0.100和0.200μmol/L尼莫地平的ACSF灌流脑片,每个浓度灌流10 min,观察RRDA的改变。结果模型第二组舌下神经根的RRDA的呼吸周期、吸气时称、放电积分幅度等呼吸指标在60 min变化差异无统计学意义;0.025μmol/L和0.050μmol/L的尼莫地平缩短RRDA的呼吸周期,0.100μmol/L的尼莫地平延长呼吸周期,且0.050μmol/L的尼莫地平能够缩短吸气时称,其余浓度无此作用,而高浓度的尼莫地平会出现过度兴奋或癫痫样放电。结论低浓度尼莫地平对RRDA有兴奋作用,高浓度则会引起神经元过于兴奋,在临床治疗中要注意药物的配伍和浓度。[Objective] To give some ideas and experimental basis for improving clinical treatment of dis- eases related to the medullary respiratory center and the use of nimodipine, the effects of nimodipine on rhythmic respiratory discharge activity (RRDA) of medullary respiration center by the use of medullary slices of newborn rat were studied. [ Methods ] Experiments were performed on medullary slice prepared from New- born rat in vitro. The slices containing the medial region of the nucleus retrofacialis (mNRF) with the hy- poglossal nerve (XII nerve) rootlets were retained. The RRDA of the XII nerve rootlets were recorded by suc- tion electrodes. The slices were perfused by Nimodipine solutions with different concentrations (0μmol/L, 0.025 μmol/L, 0.050μmol/L, 0.100 μmol/L, 0.200 μmol/L) dissolved in artificial cerebrospinal fluid (ACSF), which saturated by 95% 02 and 5% CO2 continuously, each was perfused 10 min. [Results] Nimodipine has an exciting effect on RRDA of medullaly slices in newborn rat in a low concentration, and rejection capability effect in a high concentration, even causing over excitation or epileptiform discharge. [ Conclusion] Nimodip- ine has excitatory effect on the RRDA in a low concertration, but can cause the side effect that neurons were too excited when it is in a high concentration, so the compatibility and concentration of used in clinical Ni-modipine should be cautioned.
