摘  要:活性维生素D3是一种脂溶性类固醇类衍生物,能够调节机体的钙磷代谢,在功能性食品和药品行业发挥重要的作用。利用生物转化方法将来源于植物的甾体化合物转化为活性维生素D3前体,为进一步生产维生素D3奠定基础。本研究通过摇瓶培养转化,TLC分析筛选到一株对去氢表雄酮(DHEA)转化能力较强的菌株,经形态学观察,初步鉴定为白色链霉菌。进一步通过20 L发酵罐扩大培养转化,利用硅胶柱层析、凝胶柱层析及高效液相制备柱进行分离纯化得到两种转化产物,经核磁波谱分析和旋光性测定确定了其化学结构分别为7β-OH-DHEA和7α-OH-DHEA。Activated vitamin D3, as a kind of liposoluble steroid derivatives, can be used in function food and drug industry due to its regulation to calcium and phosphate metabolism. The plant steroids such as dehydroepiandrosterone (DHEA)can be bioconverted to activated vitamin D3 precursors applied to vitamin D3 drug production. A fungus which can biotransform DHEA was isolated through the flask culture and TLC analysis, and identified as Streptomyces albus according to its morphological characteristics. The two main transformation products were purified by using silica gel column and HPLC preparation column and their structures were elucidated as 7α-hydroxy-DHEA and 7β-hydroxy-DHEA by NMR and optical activity analysis.
