摘 要:目的通过观察糖尿病大鼠视网膜血管内皮生长因子(VEGF)、蛋白激酶(PKC)的表达,探讨中药单体葛根素对于抑制糖尿病大鼠视网膜病变的作用机制。方法建立糖尿病大鼠模型,随机分为正常组(正常SD大鼠)、模型组、葛根素组、血塞通组。正常组给予正常饲养20 d;模型组给予注射等量生理盐水,10mg/kg,1次/d;葛根素组给予腹腔内注射葛根素10 mg/kg,1次/d;血塞通组给予腹腔内注射血塞通10 mg/kg,1次/d,后3组每组连续注射20 d。在第20 d过量麻醉后处死上述大鼠,轻摘眼球,进行免疫组织化学染色,检测视网膜组织中VEGF、PKC的表达,并对其进行观察比较。结果模型组PKC蛋白表达及VEGF表达均增加,葛根素组、血塞通组大鼠视网膜组织中PKC蛋白表达及VEGF表达均降低,与模型组比较差异有统计学意义(P<0.01),葛根素、血塞通两组相比,差异无统计学意义(P>0.05)。结论中药单体对糖尿病大鼠视网膜血管内皮生长因子(VEGF)、蛋白激酶(PKC)有明显的抑制作用。OBJECTIVE To discuss the mechanism of puerarin monomer to inhibit the development of diabetic retinopathy through observing the expression of the vascular endothelial growth factor(VEGF) and protein kinase(PKC) in the retina of the diabetic rats.METHODS Established the diabetic rats' model,randomly divided into model group,puerarin group,Xuesetong group and normal group(normal SD rats).The normal group was given normal breeding;The model group was injected normal saline,10 mg/kg,1 time/day;The puerarin group was injected puerarin,10 mg/kg,1 time/day;The Xuesetong group was injected Xuesetong,10 mg/kg,1 time/day.The normal group was fed for twenty days;the other groups were given continuous injection for twenty days.After twenty days put the rats to death,picked eyeballs,staining by immunohistochemica method,detected the expression of the PKC and VEGF in the retinal tissue,observed and compared the results.RESULTS The expression of the VEGF and PKC in the retina of the model group rats increased,while reduced in the puerarin group and Xuesaitong group rats.Compared with model group differences were statistically significant(P〈0.01).The differences between puerarin group and Xuesaitong group were not statistically significant(P〉0.05).CONCLUSIONS Chinese medicine monomer could inhibit the VEGF and PKC in diabetic rat's retina.
