摘  要:目的通过建立兔急性房颤的模型观察伊布利特、参松养心胶囊及两药联合用药对兔心房肌L型钙离子通道α1C、β1和钾离子通道Ikr表达的影响。方法通过建立兔急性房颤模型,应用RT-PCR、Western blot检测正常组、刺激组、伊布利特组、参松养心胶囊组和联合用药组心房肌α1C、β1、IKr基因和蛋白表达水平,并进行统计学分析。结果 (1)与正常组比较,刺激组L型钙离子通道α1C基因与蛋白表达及β1基因表达显著降低(P<0.01);而Ikr基因及蛋白表达无明显变化(P>0.05)。(2)与刺激组比较,伊布利特组α1C基因与蛋白表达及β1基因表达高于刺激组(P<0.01),Ikr基因及蛋白表达低于刺激组(P<0.01);参松养心胶囊组α1C基因与蛋白表达及β1基因表达水平与刺激组无明显差异(P>0.05),Ikr基因及蛋白表达水平无明显差异(P>0.05);联合用药组α1C基因与蛋白表达及β1基因表达高于刺激组(P<0.01),IKr基因及蛋白表达低于刺激组(P<0.01)。(3)与伊布利特组比较,伊布利特与参松养心胶囊联合用药组α1C基因与蛋白表达及β1基因表达水平无显著差异(P>0.05);Ikr基因及蛋白表达水平无显著差异(P>0.05);参松养心胶囊组α1C基因与蛋白表达及β1基因表达水平低于伊布利特组(P<0.01),Ikr基因及蛋白表达水平高于伊布利特组(P<0.01)。(4)参松养心胶囊组α1C基因与蛋白表达及β1基因表达水平低于联合用药组(P<0.01),Ikr基因及蛋白表达水平高于联合用药组(P<0.01)。结论 (1)房颤诱发术后刺激组兔心房肌L型钙离子通道α1C、β1表达明显下降,而IKr表达无明显变化。(2)伊布利特可抑制L型钙离子通道α1C、β1表达下降及降低钾离子通道IKr表达。参松养心胶囊对不能抑制L型钙离子通道α1C、β1表达下降,对钾离子通道Ikr的表达无明显影响。(3)联合用药对比单用伊布利特对α1C、β1、IKr的表达无明显影响。Objective To explore the expression of atrial myocardium's α1C、β1 and IKr in the group of Ibutilide, shensongyangxin and combined application of Ibutilide and shensongyangxin by establishing rabbit atrial fibrillation model. Methods After establishing rabbit atrial fibrillation model, used RT-PCR and Western blot to detect the mRNA and protein expression of α1C、β1and IKr in the myocardium. Results (1)The mRNA and protein expression of α1Cand the mRNA expression of β1 in stimulated group was obviously lower than the control(P 〈 0.01),and there was no significant difference of the mRNA and protein expression of Ikr between stimulated group and the control (P 〉 O.05).(2)The mRNA and protein expression of α1Cand the mRNA expression of β1 in Ibutilide group was higher than stimulated group(P 〈z 0.01), and the mRNA and protein expression of IKr in Ibutilide group was lower than stimulated group(P 〈 O.01),There was no significant difference of the mRNA and protein expression of α1C and the mRNA expression of β1 between Shensongyangxin group and stimulated group(P 〉 0.05),and there was no significant difference of the mRNA and protein expression of Ikr between Shensongyangxin group and stimulated group (P 〉 0.05). The mRNA and protein expression of α1C and the mRNA expression of β1 in combined group was higher than stimulated group(P 〈 0.01), and the mRNA and protein expression of Ikr was lower than stimulated group(P 〈 0.01).(3)There was no significant difference of the mRNA and protein expression of α1C and the mRNA expression of β1 between Ibutilide group and Combined group(P 〉 0.05),and there was no significant difference of the mRNA and protein expression of Ikr between Ibutilide group and Combined group(P 〉 0.05).The mRNA and protein expression of α1C and the mRNA expression of β1 in Shensongyangxin was lower than Ibutilide group(P 〈 0.01),and the mRNA and protein expression of Ikr was higher than Ibutilide group(P
