摘  要:目的筛选同时纯化葛根总黄酮和总皂苷的最佳吸附树脂,系统研究影响大孔吸附树脂吸附分离的多种因素,为葛根及其复方新药研发及工业化生产提供参考。方法以葛根总黄酮和总皂苷含量为考察指标,通过测定大孔吸附树脂的吸附和解析能力及吸附动力学曲线、吸附等温线、漏出曲线的绘制,优选葛根总黄酮和总皂苷的最佳纯化工艺条件。结果 D101型大孔吸附树脂纯化效果较好,其最佳工艺条件为取适当浓度的葛根提取液,上样吸附速度为2BV/h,饱和吸附后(总黄酮的饱和吸附量为54.93mg/g),先以4BV水洗脱,再分别以4BV30%、50%及70%乙醇解析。所得产物总黄酮和总皂苷含量分别为58.89%、26.32%。结论方法简便可行,纯化效果好,能为葛根复方新药研发及工业化生产提供参考。Objective To investigate the simultaneous purification of flavonoids and saponins from puerariae lobatae radix by macroporous resins,for development and industrial production of compound drug containing puerariae lobatae radix. Methods Macroporous resin was chosen with static and dynamic adsorptiion and desorption experiments to optimize the purification parameters. The contents of total flavonoids and total sapoins was taken as the index for investigation. Results D101 macroporous resin was found to have good adsorption and desorption effects.The dynamic adsorption ratio of flavonoids was 54.93 mg/g,After eluted with 4BV of distilled water and 4BV of 30%,50%,70% ethanol respectively. The contents of flavonoids and saponius were 78.89%,36.32% respectively. Conclusion D101 is a ideal resin with the best enrichment for separating and purifying total flavanoids and total saponins from puerariae lobatae radix.
